Michigan has more one-room schools than nearly any other state. Some of them are still functioning. In 1987, the Townhall School was moved from its original site on Morgan Road in Pittsfield Township to the campus of Eastern Michigan University. However, studying organ morphogenesis has been hindered by the lack of appropriate culture systems that can spatially enable cellular interactions that are needed for organ formation. Many organoid cultures rely on decellularized extracellular matrices as supportive scaffolds, which are often poorly chemically defined and allow only limited tunability and reproducibility. By contrast, engineered synthetic matrices can be tuned and optimized to mimic the embryo environment in order to enhance development and maturation of organoid cultures. Herein, this work primarily focuses on using synthetic polymer matrices to investigate how the design of biomaterials can guide key interactions guiding stem-cell decisions for the reproducible generation and control of organoid cultures.\r\nMicroporous biomaterials comprised of synthetic polymer materials were shown to guide the assembly of pancreatic progenitors into insulin-producing clusters that further developed into islet organoids. The scaffold culture facilitated cell-cell interactions enabled by the scaffold design and supported cell-mediated matrix deposition of extracellular matrix (ECM) proteins associated with the basement membrane of islet cells. Furthermore, when compared to suspension cultures, the scaffold culture showed increased insulin secretion in response to glucose stimulus indicating the development of functional \u03b2-cells. By modifying the stage that cells were seeded on scaffolds from pancreatic progenitor to pancreatic endoderm, islet organoids showed increased amounts of insulin secreted per cell. In addition, seeding scaffolds with dense clusters instead of a single suspension minimized cell manipulation during the differentiation, which was shown to be influential to the development of the islet organoids. An engineered insulin reporter further identified how mechanistic changes in vitro influenced function within individual cells by measuring insulin storage and secretion through non-invasive imaging.\r\nhPSC-derived lung organoids (HLOs) were also evaluated for in vivo maturation on biomaterial scaffolds, where HLOs were shown improved tissue structure and cellular differentiation. Investigative studies demonstrated that scaffold pore interconnectivity and polymer degradation contributed to in vivo maturation, the size of the airway structures and the total size of the transplanted tissue. Polymer biomaterials were also developed to modulate local tissue and systemic inflammation through local delivery of human interleukin 4 (hIL-4)-expressing lentivirus. Microporous scaffold culture strategies improve organoid complexity and exert fine control over the system using engineering solutions, thus, allowing the community to build more realistic organoid tools. In this thesis we focus on planning contact sequences for a humanoid robot navigating in large unstructured environments using multi-contact motion, including both foot and palm contacts. In particular, we address the two following questions: (1) How do we efficiently generate a feasible contact sequence? and (2) How do we generate contact sequences which lead to dynamically-robust motions?\r\n\r\nFor the first question, we propose a library-based method that retrieves motion plans from a library constructed offline, and adapts them with local trajectory optimization to generate the full motion plan from the start to the goal. This approach outperforms a conventional graph search contact planner when it is difficult to decide which contact is preferable with a simplified robot model and local environment information. We also propose a learning approach to estimate the difficulty to traverse a certain region based on the environment features. By integrating the two approaches, we propose a planning framework that uses graph search planner to find contact sequences around easy regions. When it is necessary to go through a difficult region, the framework switches to use the library-based method around the region to find a feasible contact sequence faster.\r\n\r\nFor the second question, we consider dynamic motions in contact planning. Most humanoid motion generators do not optimize the dynamic robustness of a contact sequence. By querying a learned model to predict the dynamic feasibility and robustness of each contact transition from a centroidal dynamics optimizer, the proposed planner efficiently finds contact sequences which lead to dynamically-robust motions. We also propose a learning-based footstep planner which takes external disturbances into account. When she retired, she began again, highlighting her culture through her art; her looking glass into the past, present and future of African-Americans with style and grace. Her work focuses on the environment of a mighty people. Working on collages allows her to blend bright colors and facial expressions that burst out of the scraps of paper that comes into its own. She works from the inside out. Though she works deliberately and consciously employing her \u201cmixed-media style,\u201d her unconscious is the undisputed movement in charge. She was a 2016 and 2017 Bombay Sapphire Regional Finalist. She also won the 2016 Spirit of the City Ford Arts, Beats, & Eats Award.
