Both are regulated by gonadal hormones, specifically estrogen and progesterone. Given that 85% of women use oral contraceptives, the most common type of hormonal contraceptives (HCs), for an average of five years, a key knowledge gap is understanding the impact of HCs on the brain. For some people, HCs increase the risk for depression, likely via the impact of HCs on stress-related responses. Nevertheless, there is little known about the mechanisms by which this occurs. The overall goal of my research is to utilize a mouse model recently developed in our laboratory to understand how HCs contribute to the vulnerability to depression. There is consistent evidence that HCs blunt hypothalamic-pituitary-adrenal (HPA) axis response to acute stress. Moreover, there is strong evidence that both estradiol and progestins (and metabolites) modify motivational processes. By using a mouse model of HC exposure to study these questions, we will be able to systematically study the impact of HCs on specific psychological and physiological components of depression, including anhedonia, motivation, and stress. We will also examine the effects of different HC formulations and determine underlying changes in circuit activation and stress-related signaling in the brain after HC exposure. Jazz, gospel, pop music with all its branches ranging from Broadway songs to hard rock via rock'n\u2019roll and funk, are some of them. But there is another stream, running through the classical music of the twentieth century. Many composers turned their attention to the blues, and the number of Jewish classical composers who wrote blues is striking. Just to name a few: Castelnuovo-Tedesco, Milhaud, Copland, Gershwin; and Ullmann and Schulhoff who perished in the Shoah. A published author and lecturer, Professor Bragalini was Distinguished Scholar at Reed College (Portland, OR) where he offered a series of lectures on Ellington. Other than potential misspecified nuisance models, most existing methods do not account for the potential problem when the variance of outcome is heterogeneous among covariates and treatment. We observe that such heteroscedasticity can greatly affect the estimation efficiency of the optimal ITR. In this presentation, we demonstrate that the consequences of misspecified treatment-free effect and heteroscedasticity can be unified as a covariate-treatment dependent variance of residuals. To improve efficiency of the estimated ITR, we propose an Efficient Learning (E-Learning) framework for finding an optimal ITR in the multi-armed treatment setting. We show that the proposed E-Learning is optimal among a regular class of semiparametric estimates that can allow treatment-free effect misspecification. In our simulation study, E-Learning demonstrates its effectiveness if one of or both misspecified treatment-free effect and heteroscedasticity exist.
