DESCRIPTION:A dozen selections from the Janice Bluestein Longone Culinary Archive from our Special Collections Research Center have been perfectly paired with dishes from the International Museum of Dinnerware Design to provide a feast for the eyes.\n\nA “perfect pairing” usually refers to a taste compatibility between wine and a food group\, such as wine and cheese. For example\, some believe a perfect pairing would be Cabernet with duck confit with turnips or Pinot Noir with bison rib eye steaks with roasted garlic — wines with sauces\, spicy food\, hors d’oeurvres\, etc. But other things can be perfectly paired such as fruit and cheese\, a couple\, or a clothing selection.\n\nSometimes pairs are made more perfect when they are catalysts for the imagination. That is what curators Margaret Carney\, Ph.D.\, and Juli McLoone are serving in \"A Perfect Pairing of Cookbooks and Dinnerware.\" The right ingredients along with an inspired recipe creates a delicious and beautiful meal that is enhanced when the cuisine is presented on a thoughtfully curated table setting\, or\, we hope\, in a thoughtfully curated exhibit. The course is both hands-on and conceptual. It begins by discussing what is unique about web surveys and when their use is most appropriate\, followed by an introduction to survey errors that can affect the quality of web survey data. Small groups of students will each develop a research problem and a questionnaire to address their problem\, designed for online administration. They will pretest the question wording\, program the questionnaire using a web survey development platform (no programming experience is required)\, and assess users’ (respondents’) experience while interacting with the web-based instrument. Students will also develop basic plans for data collection and analysis. DESCRIPTION:Analysis of Complex Sample Survey Data\n\nOpen for registration!\n\nJune 6 - July 29\, 2022\n10:00am-12:00pm\nM/W/F\n\nThis course provides practical methods and tools to analyze complex survey data with a hands-on introduction to the use of specialized statistical software procedures. The course focuses on case studies with specific large-scale national surveys: the National Comorbidity Survey-Replication (NCS-R)\, the National Health and Nutrition Examination Surveys (NHANES)\, and the Health and Retirement Study (HRS). Relevant design features of the NCS-R\, NHANES and HRS include survey weights that take into account differences in probability of selection into the sample and differences in response rates\, as well as stratification and clustering in the multistage sampling procedures used in identifying the sampled households and individuals. After introducing essential concepts related to complex sample designs\, the course will turn to the construction of survey weights\, estimation of sampling variance\, descriptive analysis\, regression analysis\, and finally special topics in the analysis of survey data. SUMMARY:Lecture / Discussion:Discovery and Development of Agonist Antibodies for T Cell Receptors
DESCRIPTION:Agonist antibodies that activate co-stimulatory immune receptors\, such as the tumor necrosis factor (TNF) receptors OX40 and CD137\, are an important class of emerging therapeutics due to their ability to regulate immune cell activity. Despite their promise\, there are no approved agonist antibodies for treating cancer as demonstrated by previous unsuccessful clinical trials. Although multiple factors are responsible for poor clinical efficacy\, one major bottleneck is the reliance on FcγR-mediated crosslinking for sufficient receptor activation. This is inherently problematic because FcγR expression varies greatly on different immune cells\, leading to a wide range of receptor agonism. Emerging research suggests that antibodies engaging two different epitopes on the same immune receptor mediate receptor superclustering and enable robust antibody agonism without extrinsic Fc crosslinking. However\, there are no systematic methods for identifying such biepitopic (also known as biparatopic) agonist antibodies. Therefore\, the objective of this research work is to develop facile methods for reliably identifying biepitopic antibodies to activate immune receptors for immunotherapeutic applications.\n\nBiepitopic antibodies have been shown to mediate potent receptor activation for a variety of immune receptors. Traditionally\, the generation of these antibodies requires key steps including animal immunization\, epitope binning to identify unique antibody pairs\, and combining antibody pairs to engineer biepitopic antibodies. While this approach has been used to successfully discover biepitopic antibodies\, it suffers from key limitations. Notably\, animal immunization and subsequent antibody isolation is an arduous and unpredictable process. Even when successful clones are discovered from these processes\, further epitope binning experiments are needed to select antibody pairs to discover potent immune therapeutics. To overcome these limitations\, we developed an antibody screening strategy that greatly simplifies the discovery of biepitopic antibodies. Our approach eliminates the need for animal immunization by using existing\, off-the-shelf IgG antibodies specific to the target receptor. Next\, we perform in vitro selections by blocking the receptor epitope of the existing antibody and conducting subsequent sorts to identify single-chain antibodies with orthogonal binding domains. Thus far\, our work has shown that the antibody screening strategy can be used to discover antibodies for a variety of TNF receptors including OX40 and CD137.\n\nGiven that receptor clustering of three or more receptors is critical for activating TNF receptors\, we first generated biepitopic tetravalent OX40 antibodies by attaching novel single-chain antibodies to the C-termini of the light chain of existing clinical-stage antibodies. These tetravalent biepitopic antibodies showed remarkable T cell proliferation and cytokine secretion for biepitopic antibodies compared to their monoepitopic counterparts. Next\, we sought to improve the additional clinical-stage OX40 IgGs engineered as biepitopic antibodies to show the generality of our findings that biepitopic antibodies can mediate superior and FcγR-independent activities. Beyond OX40 IgGs\, we also show that biepitopic antibodies can be used to mediate superior T cell proliferation for other TNF receptors including CD137. This beautifully illustrated story takes us back in time to the jungles of Mexico to discover how Mayan scholars developed a sophisticated understanding of astronomy\, architecture\, and mathematics that enabled them to predict solstices\, solar eclipses\, weather patterns and planetary movements.\n\nThe state-of-the-art Planetarium & Dome Theater at the U-M Museum of Natural History transports visitors beyond distant stars and back in time from the comfort of reclining seats. Tickets $8. Tickets are available on the day of the show at the Museum Store.\n\nThe planetarium is operating at half capacity to maximize distancing between viewers.

DESCRIPTION:Michigan songwriter Mike Vial writes songs about love and life's transitions\, and his vocal style shows the influence of James Taylor and Ari Hest. But his accomplished guitar playing has a voice of its own\, mixing elements of jazz\, folk\, and rock. Out on the road once more\, Mike is taking Mike Gentry along for the ride. Mike started out playing guitar at Dexter High School\, worked for years at A&M in Los Angeles\, and came home to Michigan. He's appeared at Plymouth Coffee Bean\, Black Fire Winery\,  Crazy Wisdom Bookstore and Tea Room\, Dearborn Brewing\, and many other venues. \"I pitched to him the idea to tour together this summer like old folk artists did in the 60s!\" Vial says. The Mikes will complete a tour of one-night stands with suitcases and guitars in hand\, with Mike performing with his band\, Chickenwire Canoe\, and ending shows as a duo. Their musical partnership feels fated\, since Gentry produced Vial's ambitious album \"A World That's Bigger.\"\n---\nProof of COVID vaccination required for entry. Open houses involve presentations on a range of fascinating astronomical phenomena\, instruction on the telescopes\, and\, when weather permits\, observing with our beautiful historic Fitz telescope as well as modern supplemental telescopes.\n\nPlease note: astronomy events at the Observatory take place even if the weather does not permit observing. We strive to always have interesting things for you to do. Tickets are required\, and open house registrations are capped at 100 guests. You can arrive anytime from 8:30 pm to 10:30 pm. You will be given a time slot for visiting the dome. While you are waiting\, check out other features and presentations.
