Known as Polskie szk\u0142o artystyczne (Polish art glass), the works were produced in glass factories in southern Poland and are a feature of many homes throughout Central Europe. The glass masters were trained in schools of art and design and many achieved international fame during their lifetimes. \r\n\r\nThe collectors, Endi Poskovic and his wife Julie Anne Visco, began acquiring the glass in 2015-16 while Endi was a Fulbright Scholar and Visiting Professor at the Jan Matejko Academy of Fine Arts in Krak\u00f3w. Scouring flea markets, antique shops, and websites, they continue to acquire pieces and build the collection to this day. We are grateful to them for making this remarkable exhibit possible at CCPS and WCEE.\r\n\r\nOrganized by the Copernicus Center for Polish Studies, this exhibition is co-sponsored by the Penny W. The exhibit also includes works of Japanese art and cartography in order to consider the dichotomy between artistry and geographic depiction, and how that plays with the definition of a \u201cmap.\u201d\r\n\r\nAlongside the exhibit, the manga map is also part of a new digital humanities preservation project at the library using the online crowd-sourcing platform Zooniverse, where the map will be transcribed\/translated and made into a fully interactive digital map. In the years immediately preceding the dam\u2019s construction, the American University in Cairo directed a large-scale project of salvage anthropology with funding from the Ford Foundation. \r\n\r\nThis endeavor yielded hundreds of photographs of al-nuba al-qadima or \u201cOld Nubia\u201d the term affectionately used by community members. Over the past sixty years, Nubians have used these images to cultivate a collective memory of a lost homeland. These ceramics act as lifeboats, which ferry the Ukrainian resistance through the flood waters of destruction. Lisova\u2019s series of tapestries explore the power of cultural memory to grow in times of war. Traditional embroidery explodes on the surface of photo collage, where images of the past and present collide on a single surface. Like a lifeline, red thread connects these projects, weaving through clay and fabric, bringing tradition to bear on new significances and the cultural will to survive. She is currently pursuing her MFA in Photography at School of the Art Institute of Chicago.\r\n\r\nIn Lee\u2019s photographic exploration, she investigates the profound connection between places and structures and the echoes of trauma that inhabit them. \u201cHolding Places\u201d is an exhibition that immerses viewers into a visual narrative, inviting them to witness the power of space as holders and conduits for personal memory.\r\n\r\nBy reconstructing these places by hand in model scale and rendering them not as they were, but how she experienced them, she is able to navigate intimate details and hidden narratives that exist within them. The process of crafting these miniatures becomes a meditative contemplation, giving Lee time to sit and reflect on these past events.\r\n\r\nThrough Lee\u2019s lens, they capture the visual manifestations of the ghosts of the past. Buckius creates \u201ctechnoffsprings\u201d: complex machines that weave together the history of inventions related to the gendered labor of women, especially regarding women\u2019s social roles as caregivers and subjects of care themselves. \r\nTrained as an engineer and an artist, Buckius\u2019 machines are intentionally complex, layered, and illogical or absurdly logical. In the nature of women\u2019s caregiving, they teeter between order and chaos. Her \u201cdigital tinkerings\u201d tell epic tales of motherhood, technology, female bodies, and commerce\u2014both personal and externalized through women\u2019s inventions and early forays that bridged caregiving and commerce. One of the major factors of increasing rates of antibiotic resistance is that most antibiotics target the bacterial ribosome or a limited range of proteins. Due to the narrow range of targets and the relatively small number of antibiotics, resistance to commonly used treatments arises quickly and can spread across bacterial populations. While there have been efforts to identify new targets for antimicrobial compounds and develop antibiotics with different chemical structures and mechanisms of action, there continues to be a dearth of innovation in developing truly innovative antibiotic compounds. \r\n\r\nIn this thesis, I discuss efforts to develop a series of tools to aid in the identification of new molecular scaffolds, small molecules that can be altered to optimize antibacterial effects, for an underexplored antibiotic target. Specifically, these methods target bacterial riboswitches, structured RNA elements that regulate vital biosynthetic pathways by binding a specific small molecule. Riboswitches are present in many bacteria, including bacteria that pose the greatest risks to human health. Each riboswitch is defined by the specific molecule it binds to; this dissertation focuses on the flavin mononucleotide riboswitch (FRS) The FRS binds to flavin mononucleotide, an important cofactor for protein function and a key part of multiple metabolic cycles. Bacteria often use FRS to regulate riboflavin biosynthesis, which is crucial for cellular growth and energy production. Since the FRS is a regulator for a vital pathway, identifying molecules with distinct molecular structures that bind to the riboswitch could be optimized into novel antibiotics. \r\n\r\nWhile FRS has a wealth of previous characterization and some previously identified binding partners; however, none of these candidates have advanced beyond preliminary mouse studies, far from actual implementation as a drug. We developed Fluorescent Ligand Equilibrium Displacement (FLED), a robust, rapid, and repeatable method, to identify molecules capable of interacting with the FRS. This method was applied to large libraries of compounds and their preliminary hits, investigated using classical biochemical techniques. We also pioneered methods for analyzing RNA using the analytical technology Ion Mobility Mass Spectrometry (IM-MS). We utilized it to study both transfer RNAs and FRS. While the previously mentioned methods are versatile tools for screening compound libraries, they do not account for the high cost of molecule libraries, nor the limitations inherent to screening vast molecular libraries. We employed two computational screening methods to reduce the number of compounds screened before identifying a new binding partner. These methods, coupled with FLED, were used to investigate the predictions and determine if the current setup does increase the rate of compound discovery.\r\n\r\nThe results described here support using FLED, IM-MS, and computational screening to identify small molecule candidates for development as FMN riboswitch-targeted antibiotics. It also provides preliminary work toward new a method to measure transcription termination of riboswitches.
