DESCRIPTION:David Shechner Ph.D.\nAssistant Professor of Pharmacology\nUniversity of Washington\n\nKeywords: RNA\, proximity-biotinylation\, subcellular architecture\, nuclear architecture\, spatial biology\, biomolecular condensates\, interactomics\, nucleolus\, Xist.\n\nAbstract: In the context of the living cell\, very little RNA is naked. RNA molecules form complex\, dynamic networks of molecular interactions that underlie a host of biochemical functions\, and which are central to organizing subcellular compartmentalization. In humans\, for example\, RNAs are key determinants of chromatin folding\, and they nucleate and scaffold a host of biomolecular condensates that collectively control cellular metabolic\, epigenetic\, and stress-signaling pathways. But\, characterizing these structures—identifying the biomolecules within an RNA's subcellular microenvironment—remains technically cumbersome. \n\nTo address this challenge\, I introduce oligonucleotide-mediated proximity-interactome mapping (O-MAP)\, a straightforward and flexible method for identifying the proteins\, RNAs\, and genomic loci near a target RNA\, within its native cellular context. O-MAP uses programmable oligonucleotide probes to deliver proximity-biotinylating enzymes to a target RNA. These enzymes then pervasively label all nearby (~20 nm) molecules\, enabling their enrichment by streptavidin pulldown. O-MAP induces exceptionally precise RNA-targeted biotinylation\, and its modular design enables straightforward validation of probe pools and real-space optimization of the biotinylation radius\, thus overcoming key technical challenges for the field. Moreover\, O-MAP can be readily ported across different target RNAs and specimen types\, including patient-derived organoids and tissue samples. And\, O-MAP achieves this without complex cell-line engineering\, using only off-the-shelf parts and standard manipulations. \n\nUsing a small cohort of model RNAs\, we have developed a robust O-MAP toolkit for proteomic (O-MAP-MS)\, transcriptomic (O-MAP-Seq) and genome interaction (O-MAP-ChIP) discovery. O-MAP of the 47S-pre-rRNA—the long noncoding RNA that scaffolds the nucleolus—enabled a comprehensive \"multi-omic\" analysis of this subnuclear structure\, and revealed hundreds of novel nucleolar protein-\, RNA-\, and chromatin interactions. O-MAP of XIST—the master regulator of X-chromosome inactivation—revealed novel RNAs that may play a role in this process\, and unanticipated interactions between XIST and other chromatin-regulatory RNAs. Finally\, targeting O-MAP to introns within a key cardiac developmental gene enabled unprecedented molecular dissection of a subnuclear compartment that would be impossible to purify biochemically.\n\nGiven these results\, we believe that O-MAP will be a powerful tool for elucidating the mechanisms by which RNA molecules drive subcellular compartmentalization in time and space\, with particular impact on our understanding of nuclear architecture. DESCRIPTION:This paper estimates the effects of large donors’ political contributions on charitable contributions and fundraising. Political contributions reduce the effectiveness of charitable fundraising\, particularly in the Health and Other sectors. Charities therefore reduce fundraising\, which contributes to an overall elasticity of private charitable contributions to political contributions of -0.07. This aggregate effect combines large political donors’ charitable contribution behavior with spillover effects of their political giving on the charitable giving patterns of the remainder of the population. Subsequent analysis allows for heterogeneity in the ideologies of both political and charitable recipients.

DESCRIPTION:Speaker: Kevin Smith\, PhD Candidate\, U-M IOE\n\nDescription: Social Inequality can be defined by unequal outcomes that occur as a result of uneven distribution of resources. Many of the disparities that are observed across income quantiles occur in concentrated United States communities. Within these concentrated areas\, unique social and behavioral responses exist to healthcare\, health policy\, and public health policy. While the US collects an enormous amount of demographic\, health\, and economic data at various community levels\, there still exists a gap in how decision-makers can leverage these data to design appropriate community policy initiatives whose outcomes are intended to reduce the observed disparities nationwide. In this seminar\, we will discuss applications of machine learning models in public health and healthcare policy and challenges in their respective outcome predictions.\n\nBio: Kevin B. Smith is a PhD candidate in the department of Industrial and Operations Engineering (IOE) at the University of Michigan and is co-advised by Dr. Brian Denton and Dr. Siqian Shen of IOE. His research is focused on applying industrial engineering and operations research tools to improve outcomes resulting from problems of social inequality. The mission of his research is to inform policymakers and the public of the critical factors that impact decisions with particular applications in organ allocation\, pandemic risk planning\, and clinical kidney stone care thus far. Additionally\, he is motivated to allocate resources equitably and improve decision-making outcomes in these settings. DESCRIPTION:Sign up for a free series of self-defense workshops!\n\nAiming to equip the incoming students with the skills to increase their self-confidence and preparedness for the unexpected\, the Engineering Student Government (ESG) in partnership with UofM Division of Public Safety & Security (DPSS) is setting out with a newly established SaferSelf project. Starting on September 12th and running until October 10th a series of free empowerment self-defense workshops will be held weekly on Mondays from 5:30pm to 6:30pm on Central Campus (exact location TBD).\n\nEmpowerment self-defense sessions explore the culture of violence\, and teach concrete but practical effective skills for personal safety and physical self defense in a wide variety of contexts ranging from harassment to sexual assault. Participants will be able to:\n- understand how situational awareness can deter or prevent an attack.\n- use verbal skills for assertive communication.\n- evade and set boundaries (verbal & physical).\n- use practical options to make ourselves more comfortable when uncomfortable situations occur.\n- recognize and interrupt unwanted behavior when in social situations\, interpersonal/intimate relationships as well as interactions with strangers.\n\nThe series will be composed of the following workshops:\n1. Introduction to physical self-defense: 1.5hr\n2. Physical self-defense with strikes and kicks with pads: 1hr\n3. Physical self-defense with boxing element combination striking: 1h\n4. Top-to-bottom pressure points: 1hr\n5. Physical self-defense from the ground: 1hr\n\nFace masks will be required regardless of vaccination status.\n\nThe space in the program is limited\, and last spots are available. DESCRIPTION:A dozen selections from the Janice Bluestein Longone Culinary Archive from our Special Collections Research Center have been perfectly paired with dishes from the International Museum of Dinnerware Design to provide a feast for the eyes.\n\nA “perfect pairing” usually refers to a taste compatibility between wine and a food group\, such as wine and cheese. For example\, some believe a perfect pairing would be Cabernet with duck confit with turnips or Pinot Noir with bison rib eye steaks with roasted garlic — wines with sauces\, spicy food\, hors d’oeurvres\, etc. But other things can be perfectly paired such as fruit and cheese\, a couple\, or a clothing selection.\n\nSometimes pairs are made more perfect when they are catalysts for the imagination. That is what curators Margaret Carney\, Ph.D.\, and Juli McLoone are serving in \"A Perfect Pairing of Cookbooks and Dinnerware.\" The right ingredients along with an inspired recipe creates a delicious and beautiful meal that is enhanced when the cuisine is presented on a thoughtfully curated table setting\, or\, we hope\, in a thoughtfully curated exhibit.
