SUMMARY:Workshop / Seminar:MCDB Seminar> How Dynein Teams Adapt to Load and Coordinate Their Motors
DESCRIPTION:Cytoplasmic dynein drives long-distance transport along microtubules\, but how motor complexes adapt to mechanical loads and coordinate their activities remains poorly understood. I will present our discovery that the dynein–dynactin–BicD2 (DDB) complex dynamically recruits a third dynein under load through an auxiliary BicD2 adaptor that interacts with the dynein light intermediate chain (LIC)\, increasing force generation from 7 to 9 pN. I will also show how our investigation of the neurodevelopmental disease-associated DYNC1H1 mutation P3018S revealed that the trailing dynein determines the velocity of multi-dynein complexes\, while the leading dynein stimulates the trailing motor through its LIC. Together\, these findings uncover unexpected roles for LICs in load-dependent motor recruitment and inter-motor communication\, establishing new principles of dynein force adaptation and coordination.\n\n\nThe Gennerich Lab investigates the molecular mechanisms by which biomolecular motors generate force and motion in eukaryotic cells. We focus on understanding how defects in these processes contribute to human disease. Our research combines single-molecule biophysics\, molecular biology\, and biochemistry to study the function and regulation of microtubule-based motors—kinesins and cytoplasmic dynein—as well as RNA polymerase II during transcription elongation through chromatin.\n\nUsing a multidisciplinary approach that includes high-resolution optical trapping\, single-molecule fluorescence microscopy\, and CRISPR-Cas9-based genetic tools\, we dissect how these motor proteins convert chemical energy into mechanical work and how their activity is modulated in different cellular contexts. Our long-term goal is to uncover the design principles of biomolecular motors and to elucidate the molecular basis of diseases caused by impaired motor function.

SUMMARY:Lecture / Discussion:'Auditing AI' Book Launch & Reception
DESCRIPTION:The Center for Ethics\, Society\, and Computing is delighted to invite you to celebrate the book launch of Auditing AI with authors Christian Sandvig\, Ellery Biddle\, and Danae Metaxa! \n\nTime: Nov. 9\, 4 pm – 5 pm (Authors Discussion)\, 5 pm -7 pm (Reception with appetizers\, beer and wine)\n\nLocation: Cahoots Event Space (206 E Huron St)\n\nPlease RSVP: https://tinyurl.com/2wa4r3up\n\nAbout the book:\n\nHow tech companies\, journalists\, and policymakers can prevent AI decision-making from going wrong.\n\nOur lives are increasingly governed by automated systems influencing everything from medical care to policing to employment opportunities\, but researchers and investigative journalists have proven that AI systems regularly get things wrong.\n\nAuditing AI is a first-of-its-kind exploration of why and how to audit artificial intelligence systems. It offers a simple roadmap for using AI audits to make product and policy changes that benefit companies and the public alike. The book aims to convince readers that AI systems should be subject to robust audits to protect all of us from the dangers of these systems. Readers will come away with an understanding of what an AI audit is\, why AI audits are important\, key components of an audit that follows best practices\, how to interpret an audit\, and the available choices to act on an audit’s results.\n\nThe book is organized around canonical examples: from AI-powered drones mistakenly targeting civilians in conflict areas to false arrests triggered by facial recognition systems that misidentified people with dark skin tones to HR hiring software that prefers men. It explains these definitive cases of AI decision-making gone wrong and then highlights specific audits that have led to concrete changes in government policy and corporate practice.\n\nAbout the ESC:\nThe Center for Ethics\, Society\, and Computing (ESC – pronounced “escape”) is dedicated to intervening when digital media and computing technologies reproduce inequality\, exclusion\, corruption\, deception\, racism\, or sexism. ESC is generously supported by the School of Information\; the Center for Political Studies at the Institute for Social Research\; and the Department of Communication & Media in the College of Literature\, Science\, and the Arts at the University of Michigan.

SUMMARY:Lecture / Discussion:Dynamics seminar: Dynamics of the Secant Method and Other Monomial-Type Maps
DESCRIPTION:We discuss the secant method\, a root-finding algorithm alternative to Newton’s method. Given a rational map f : P^1 -> P^1\, the secant method yields a dynamical system Sf on P^1 x P^1 (minus a finite indeterminacy locus). If r in C is a simple root of f\, then the point (r\, r) is an attracting fixed point of Sf.\n\nAdditionally\, when 0 is a simple root of f with f’’(0) nonzero\, we have that Sf is an instance of the broader class of “monomial-type maps.” A “monomial map” is simply a function whose output coordinates are all monomials in the input coordinates. A “monomial-type map” is then a coordinate-by-coordinate scaling of a monomial map by some holomorphic scalar functions. In the aforementioned case for 0 and Sf\, we find that Sf(x\, y) = (a(x\, y) * xy\, x). \n\nBy extending the 1-dimensional constructions of Böttcher coordinates and Green’s functions to these higher-dimensional monomial-type maps\, we can show that (1) Sf has well-defined dynamical potential\, (2) it takes the vequipotential to the v^φ equipotential (where φ is the golden ratio)\, and (3) Sf is locally modeled by the Fibonacci monomial map (x\, y) |-> (xy\, x)\, whose dynamics are well-understood.
