H. Waddington Professor of Pediatrics at Harvard Medical School. I received my PhD from New York University and postdoctoral training at Princeton University. I joined Harvard Medical School as an Assistant Professor in 1991 and was appointed a Professor of Pathology in 2004. In 2009 I joined the Newborn Medicine Division of Boston Children’s Hospital.\n\nI am interested in identifying key epigenetic regulators in cancer\, elucidating their mechanism of action and providing the conceptual basis for translating our basic findings to the clinic via the development of new therapeutic strategies. With the discovery of the first histone methyl eraser\, LSD1\, in 2004\, our group demonstrated that histone methylation is dynamically regulated\, which overturned the long-held dogma that such modifications were static and irreversible. We have also discovered many additional histone demethylases with different specificities\, and novel readers\, including those that specifically recognize unmodified lysine and arginine and suggest that the unmodified states are not simply a ground neutral state of epigenetic information but rather likely code for epigenetic information as modified states. Importantly\, many of these chromatin enzymes and readers have since been implicated in various types of human cancers\, indicating an important role of chromatin regulation in tumorigenesis.\n\nMore recently\, we have also been studying RNA modifications and how they impact gene expression regulation. In many ways this exciting field parallels the early days of chromatin biochemistry and biology\, i.e.\, the nature and the biological and pathological functions of RNA modifications\, as well as the enzymes responsible for writing\, erasing and reading them\, are just beginning to be understood.\n\nAt Ludwig Oxford\, my lab is focusing on two questions. First\, how to convert “cold tumors to “hot” and how to sustain durable responses to cancer immune checkpoint blockade therapy. Second\, how to induce therapeutic differentiation of cancers\, using acute myeloid leukemia and diffuse intrinsic pontine glioma as models where chromatin/epigenetics have been shown to play a crucial role in the maintenance of a poorly differentiated state. It has been running uninterruptedly since it was founded by Kurt Lewin in the 1920’s in Berlin. A very important feature of this seminar today is its interdisciplinary nature. Some might even call him a “tour de force” as his songs pull no punches in evoking the demons with which he has wrestled through much of his life. \n\nThe GRAMMY-nominated multi-platinum singer/songwriter’s early releases\, I’m About To Come Alive\, The Sound Of A Million Dreams\, I’m A Fire and Fighter\, ignited his reputation as an innovator and creative risk-taker yet left Nail feeling restless. This led to the departure of the only record company home he’d ever known\, MCA Nashville\, where he’d formed friendships that endure even now. Through dealing with his own journey of mental health and the feeling of needing to “rebel against what people expect you to be\,” Nail began making his most “organic” work to date\, David Nail & The Well Ravens\, with Jason Hall and Andrew Petroff. That project liberated Nail and propelled him into his next adventure. Writing solo for the next few months led to his debut solo independent EP Oh\, Mother\, a reflective piece of work. Of the title track\, Rolling Stone noted that “the song creeps in like a sunrise — quiet\, restrained cello and keys are the only underpinning for Nail’s powerful vocals at first. But then it slowly builds to something more massive and majestic.” That release was followed by Bootheel 2020 and Bootheel 2021\, projects dedicated to the singer/songwriter’s hometown in the “bootheel” of Missouri. He received a B.A. in Integrative Biology from the University of California at Berkeley (1998)\, and worked as a technician for 2 years in Sarah Hake's plant genetics group at the USDA/ARS Plant Gene Expression Center. He received his Ph.D. in genetics from Cold Spring Harbor Laboratory (2003)\, where his work in Shiv Grewal's laboratory established the first direct link between RNA interference and chromatin-based epigenetic inheritance. As a postdoc with Michael Wigler (2004) and independent Cold Spring Harbor Laboratory Fellow (2004-2007)\, Dr. Hall used microarray technologies and mouse strain genealogies to conduct the first systematic study of DNA copy number variation hotspots. As a faculty member at the University of Virginia (2007-2014)\, Washington University (2014-2020) and Yale (2020-present)\, his work has sought to understand the causes and consequences of genome variation in mammals\, with an increasing focus on computational methods development and human genetics. His group has developed bioinformatics tools for variant detection\, variant interpretation\, sequence alignment\, data processing\, and data integration. He has led genome-wide studies of human genome variation\, heritable gene expression variation\, human genetic disorders\, tumor evolution\, mouse strain variation\, genome stability in reprogrammed stem cells\, and single-neuron somatic mosaicism in the human brain. Dr. Hall's work has been featured in Science Magazine's Breakthrough of the Year (2003 & 2007)\, the NIMH Director's \"Ten Best of 2013\" and The Scientist (2013)\, and he has received several prestigious awards including the AAAS Newcomb Cleveland Prize (2003)\, the Burroughs Wellcome Fund Career Award (2006)\, the NIH Director's New Innovator Award (2009)\, and the March of Dimes Basil O'Connor Research Award (2010). He has also served as an Associate Editor at Genome Research (2009-2014) and Genes\, Genomes and Genetics (2011-2018).\n\nMost recently\, Dr. Hall has played a leadership role in several large collaborative projects funded by NIH/NHGRI including the Centers for Common Disease Genomics\, the AnVIL cloud-based data repository and analysis platform\, and the Human Pangenome Project. His current work is focused on two broad goals: (1) mapping variants and genes that confer risk to human disease\, with ongoing projects focused on coronary artery disease and cardiometabolic traits in unique and underrepresented populations\, and (2) developing methods for the detection and interpretation of human genome variation\, with an emphasis on structural variation and other difficult-to-detect forms\, and on comprehensive trait association in human disease studies. In the wake of the international order’s dismantling and the ruinous acceleration of techno-capitalism\, social discontent has been channeled into extreme nationalisms that promise a ‘homecoming’ at the expense of outsiders\, immigrants\, refugees. Adding to these woes\, intellectual efforts to challenge the ‘dogmatic images’ of thinking that have ruled by elevating particular epistemologies to the level of ‘global thought\,’ are being increasingly stymied by capitulant institutions. Yet\, it is precisely in such precarious times that cross-cultural thinking seems more necessary than ever\, not least as an antidote to the ongoing segregation of knowledges and thought-systems currently underway.\n \nThis conference is part of a series of events organized under the Geophilosophies initiative. As a project Geophilosophies not only calls attention to thought-systems that continue to be erased\, discredited\, or ignored by dominant cultures\, but more importantly\, encourages the kind of deep conceptual encounter between different traditions of thought with the capacity to effect real psychic\, social and ecological change. We define cross-cultural thinking as the kind of engagement that strives for maximum communication\, unlike the concept of ‘border crossing’ which we see as a vestige of nation-state sovereignty. Thus defined\, cross-cultural thinking is transversal and ecosophical by nature\, inasmuch as it strives for modes of thinking grounded in lived practices and experiences that effortlessly bring incompatible concepts and entities into new associations characterized by their mutual self-transformation. On this note\, we invite presentations that re-examine unexplored avenues in cross-cultural philosophical thinking\, aiming to repair fractured connections between self\, society and world amid today’s global turmoil. We are particularly interested in experimental methodologies that explore: (i) the nature of contact zones between concepts\, praxes\, and worldviews\; (ii) the co-imbrication of one’s subjectivity in cross-cultural encounters. We encourage risk-taking\, for example\, by resisting the kind of knowledge specializations that may prevent genuine engagement between differences and that work through two or more traditions of thought to create an ‘in-between’ idiom that is at home in its homelessness.\n \nPursued in collaboration with the Uberoi Foundation\, an organization that is dedicated to cross-cultural exchange and expanding awareness of diverse traditions\, this conference promises to break new ground in the development of ways of thinking that simultaneously draw upon ancient human wisdoms while also being informed by the urgency of the present global moment\, bringing a diasporic consciousness to a world seduced by the alluring false comfort of restrictive nationalisms. In unconstrained problems with exact gradients\, momentum-based methods—most notably Nesterov’s accelerated gradient descent (AGD) and Polyak’s heavy-ball (HB) method—achieve faster convergence by improving dependence on the condition number. However\, this acceleration comes at a cost: momentum amplifies gradient noise\, making these methods less robust than GD under standard parameter choices and requiring more accurate gradient estimates to attain comparable accuracy. Similar challenges arise in convex and nonconvex min–max optimization.\nMotivated by applications in machine learning\, this talk studies unconstrained and min–max optimization under deterministic\, unbiased stochastic\, and biased stochastic gradient noise. I will present new algorithms that achieve optimal robustness against different noise types\, using control-theoretic tools such as the H_2​ norm\, the H_∞​ norm\, and the risk-sensitivity index\, together with coherent risk measures. I will also discuss worst-case noise constructions and high-probability convergence guarantees.
