Current paradigms for drug safety evaluation are costly, lengthy, and conservative, and impede efficient drug development. Here we combine multiscale experiment and simulation, high-performance computing, and machine learning to create an easy-to-use risk assessment diagram to quickly and reliable stratify the pro-arrhythmic potential of new and existing drugs. We capitalize on recent developments in machine learning and integrate information across ten orders of magnitude in space and time to provide a holistic picture of the effects of drugs, either individually or in combination with other drugs. We show, both experimentally and computationally, that drug-induced arrhythmias are dominated by the interplay of two currents with opposing effects: the rapid delayed rectifier potassium current and the L-type calcium current. Using Gaussian process classification, we create a classifier that stratifies safe and arrhythmic domains for any combinations of these two currents. We demonstrate that our classifier correctly identifies the risk categories of 23 common drugs, exclusively on the basis of their concentrations at 50% current block. Our study shapes the way towards establishing science-based criteria to accelerate drug development, design safer drugs, and reduce heart rhythm disorders.\r\nBio:\r\nEllen Kuhl is the Walter B. Reinhold Professor in the School of Engineering and Robert Bosch Chair of Mechanical Engineering at Stanford University. She received her PhD from the University of Stuttgart in 2000 and her Habilitation from the University of Kaiserslautern in 2004. Her area of expertise is Living Matter Physics, the design of theoretical and computational models to simulate and predict the behavior of living systems. Ellen has published more than 200 peer-reviewed journal articles, edited two books, and published a textbook on COVID-19. She is a founding member of the Living Heart Project, a translational research initiative to revolutionize cardiovascular science through realistic simulation with 400 participants from research, industry, and medicine from 24 countries. Ellen is the current Chair of the US National Committee on Biomechanics and a Member-Elect of the World Council of Biomechanics. She is a Fellow of the American Society of Mechanical Engineers and of the American Institute for Mechanical and Biological Engineering. She received the National Science Foundation Career Award in 2010, was selected as Midwest Mechanics Seminar Speaker in 2014, and received the Humboldt Research Award in 2016 and the ASME Ted Belytschko Applied Mechanics Award in 2021. And yet, we do not recognize or call out the responsibility that comes with that power. This session is meant to challenge future business leaders to think differently about their career, its purpose, and its value as a calling or vocation, one that is in service to society. Its message is for current and prospective business students, business leaders thinking anew about the role of business in society, and the business educators that train all these people. \r\n\r\nWe face great challenges as a society today, from environmental problems like climate change and habitat destruction, to social problems like income inequality, unemployment, lack of a living wage, and poor access to affordable health care and education. Solutions to these challenges must come from the market (as comprised of corporations, the government, and nongovernmental organizations, as well as the many stakeholders in market transaction, such as the consumers, suppliers, buyers, insurance companies, and banks), the most powerful institution on earth, and from business, which is the most powerful entity within it. \r\n\r\nThough government is an important and vital arbiter of the market, business is the force that transcends national boundaries, possessing resources that exceed those of many nations. Business is responsible for producing the buildings that we live and work in, the food we eat, the clothes we wear, the forms of mobility we employ, and the energy that propels us. This does not mean that only business can generate solutions or that there is no role for government, but with its unmatched powers of ideation, production, and distribution, business is positioned to bring the change we need at the scale we need it. Without business, the solutions will remain elusive. Indeed, if there are no solutions coming from the market, there will be no solutions. And without visionary and service-oriented leaders, business will never even try to find them. \r\n\r\nAbout Hoffman: \r\n\r\nAndy Hoffman is the Holcim (US) Professor of Sustainable Enterprise at the University of Michigan; a position that holds joint appointments in the Stephen M. Ross School of Business and the School for Environment and Sustainability. Professor Hoffman's research uses organizational behavior models and theories to understand the cultural and institutional aspects of environmental issues for organizations. \r\n\r\nHe has published over 100 articles\/book chapters, as well as 18 books, which have been translated into five languages. In this work, he focuses on the processes by which environmental issues both emerge and evolve as social, political, and managerial issues, including: the evolving nature of field level pressures related to environmental issues; the corporate responses that have emerged as a result of those pressures, particularly around the issue of climate change; the interconnected networks among non-governmental organizations and corporations and how those networks influence change processes within cultural and institutional systems; the social and psychological barriers to these change processes; and the underlying cultural values that are engaged when these barriers are overcome. Collins will share examples of collecting data using WhatsApp audio responses to understand changes in gender norms and assessing Google Play reviews of digital lending apps in India. In both examples, Collins will share how she and her team used Natural Language Processing (NLP) \u2013 a common market tool - to detect key topics and phrases to unpack the meaning of the responses. Collins\u2019 work is grounded in a deep understanding of the financial lives of individuals. She is the author of the ground-breaking \u201cPortfolios of the Poor\u201d and creator of the Financial Diaries, a research tool used in over 10 countries, including South Africa, Kenya, Mozambique and Tanzania.\u00a0\r\n\r\nPeople living at or near poverty levels are less likely to speak their minds when responding to surveys and other efforts to collect data. This hesitancy is understandable, but it also creates a real challenge for researchers, enterprises and policymakers to better understand their needs, said Heather Esper, Director of WDI\u2019s Performance Measurement & Improvement group.\r\n\r\n\u201cDaryl Collins is truly a pioneer in collecting and analyzing data to better understand the lives of low-income and vulnerable individuals at scale,\u201d Esper said. \u201cThe qualitative methods she uses shed more light on what individuals are saying by accounting for how they say it.\u201d\r\n\r\nCollins recently established Decodis with a team of linguists and Natural Language Processing data scientists. Decodis seeks to advance scale and robustness in qualitative research techniques by providing tech-led consumer research methods that are insightful, scalable and low cost. The Decodis team is currently working on a range of projects across a breadth of countries, languages and sectors to explore new ways of both collecting and analyzing open-end response data.\u00a0\r\n\r\nCollins holds bachelor\u2019s and master's degrees in economics from the London School of Economics and a Ph.D. from New York University.
