“Structural remodeling and volumetric growth in the heart”

Abstract:

Many cardiac diseases impose pressure overload leading to ventricular enlargement via the growth of cardiac myocytes and remodeling of the collagen fiber architecture. One such disease is pulmonary arterial hypertension (PAH), which primarily affects the right ventricle (RV). The effects of PAH on the functional behavior of the RV free wall (RVFW) on RV organ-level cardiac function remain largely unexplored. Computational rodent heart models (RHM) of the normal and hypertensive states can be quite valuable in gaining insights into the pathophysiology of myocardium remodeling. Recently, we developed high-fidelity biventricular finite-element RHMs using extensive data collected from rat hearts at normal and post-PAH time points. In this study, we extended our model to predict the time-course adaptations of RV under PAH from normal state to post-PAH state. Our model accounted for growth processes driven by sarcomerogenesis and collagen fibrosis, coupled with myo- and collagen fiber reorientation events. We used our model to investigate the correlations between the alterations in the wall stress, the remodeling of the RVFW microstructure, and the shape changes in the RV during the development of PAH. To the best of our knowledge, this is the first work to investigate the interaction between fiber remodeling and volumetric growth in the heart. Ultimately, the detailed description of organ-level remodeling patterns predicted by in-silico models such as ours could replace the traditional measures of RV dimensions and volume that often lead to gross and limited information on cardiac performance. Future directions of the research will also be presented.

“Multi-Omics Systems Biology Approach to
Vascular Biology”

Abstract:

Over the past decade, we have investigated the mechanisms associated with shear stress response in endothelial cells using systems biology approaches [e.g. 1-8]. In the most recent granting period, we have carried out longitudinal analysis of distinctive shear stress response to pulsatile and oscillatory stress corresponding to normal and disturbed flow in blood vessels. Our studies have provided fundamental insights into temporally regulated mechanisms as well as a systems-level view of stress response. Our key findings also included implications of long noncoding RNA molecules – LEENE and LINC341 - which appear to have dramatic consequences on endothelial homeostasis. The regulation by LINC RNAs entails the coupling between KLF4 and genes involved in endothelial function. In addition, we explored the interplay between endothelial cells and macrophages in atherogenesis. These studies reveal a novel mechanisms in which the transcription factor KLF4 regulates the enzyme cholesterol-25-hydroxylase and the nuclear hormone receptor Liver X Receptor. Our key findings indicate that KLF4 transactivates cholesterol-25-hydroxylase and liver X receptor, thereby promoting the synergistic effects between ECs and macrophages to protect against atherosclerosis susceptibility.

Synthetic chromosomes have the potential to revolutionize gene therapy, because they can deliver entire genetic programs. However, this strategy can succeed only if the synthetic chromosomes are faithfully transmitted to each daughter cell upon cell division. Eukaryotic cells use the kinetochore, an immensely complex protein machine, to segregate their native chromosome during cell division. We are initiating an effort to engineer a synthetic kinetochore to faithfully segregate synthetic chromosomes. This ambitious goal encompasses three aims. First, we will use bottom-up reverse engineering to yield deep insight into function and regulation of the kinetochore, potentially exposing unexpected gaps in our understanding. Second, we will identify functionally necessary kinetochore components from those that are its ‘evolutionary legacy’. This is necessary to reveal strategies for simplifying the design of the synthetic kinetochore. Third, we will establish the blueprint for designing a synthetic, orthogonal segregation machine that specifically controls the inheritance of synthetic chromosomes.

Abstract
For many years now, my students and I have developed several solutions to enable cost-effective deployments in the cloud of low-latency highly-available services. Based on this work, in this talk, my goals are two-fold.

First, I will give an overview of how some of my group's work, as well as other related research from computer science researchers, can be leveraged to enable cloud-driven control of manufacturing devices, i.e., how CS research can be used for improved manufacturing.

Second, in the context of smart manufacturing, I will describe the challenges in applying the approaches traditionally used by computer science researchers. In particular, I will argue that many of the principles that underlie use of the cloud will need to be rethought, and the domain of smart manufacturing presents an exciting new frontier for distributed systems research. I will then show some preliminary work done in collaboration with Prof. Okwudire on the application of cloud computing to the control of manufacturing machines.

Bio
Harsha V. Madhyastha is an Associate Professor in Computer Science and Engineering at the University of Michigan. His research interests span all facets of scalable, fault-tolerant, performant, and privacy-preserving networked systems. His work has resulted in award papers at several top-tier conferences and also received the IRTF's Applied Networking Research Prize on multiple occasions. He is the recipient of Google Faculty Research awards, a NetApp Faculty Fellowship, a Facebook Faculty Award, and an NSF CAREER award.

“Radically Simplifying hMSC Manufacturing Scale-up and Regenerative Medicine Product Development (and Building a Business around Regenerative Medicine Manufacturing Technology)”



Abstract:

Human Mesenchymal Stem/Stromal Cells (hMSCs) and extracellular vesicles (hMSC-EVs) are leading platforms for development of regenerative medicine therapies. Multiple bottlenecks exist during discovery, product development, manufacturing process development, and cGMP manufacturing leading to timelines from discovery through first in man testing often averaging 7-11 years. RoosterBio has developed and commercialized novel hMSC bioprocess systems that are designed for rapid process development and cGMP tech transfer to radically reduce development timelines to as little as 2-4 years. Requirements for implementation of scalable bioreactor production systems will also be addressed, and data will be presented from 50L scale microcarrier-based processes with CQAs for both hMSCs and EVs.

By stimulating or recording electrical activity, microelectrode arrays implanted in the brain have created a renaissance in the treatment of neurological diseases and injuries. Likewise, these devices are an enabling technology to understand normal brain function and behavior. However, questions remain regarding the relationship between the biological response to implanted electrodes, their chronic performance, and features of their design. It is my lab’s goal to understand the basic science underlying the interaction between implanted electrodes and brain cells, and to provide guiding principles to improve device design and performance as a result. Recently, we have found novel effects of implanted silicon and polyimide-based electrode arrays on the structure and function of local neurons, including alterations in ion channel expression, synaptic transporter expression, dendritic spine density, and excitability. Results of quantitative immunohistochemistry demonstrate a progressive local increase in the expression of potassium ion channels and inhibitory transporters surrounding devices implanted in the brains of rats over time, indicating a potential shift toward hypoexcitability over the 6-week time course studied. Two-photon laser scanning microscopy in brain slice preparations revealed profound local spine loss surrounding implants, coupled with observations of reduced responsiveness to injected current during whole cell intracellular recordings, where preliminary observations indicate particularly pronounced effects surrounding silicon-based devices. More recently, we have pursued RNA-sequencing to understand the molecular identity and function of neurons and non-neurons surrounding implanted electrodes. Our results suggest a novel role of local plasticity surrounding devices in chronic signal loss and instability, and we are currently working to assess and perturb local gene expression to reveal potential underlying mechanisms.

Photoacoustic imaging (PAI) gained significant attention of biomedical community as it provides optical
absorption contrast based functional and molecular information of very deep biological tissue at ultrasonic
resolution. In the last two decades, PAI evolved as a multi-scale imaging technology, enabling in vivo imaging from organelles to organs, and translated to several clinical applications such as breast and thyroid imaging. Nevertheless, the development of PAI systems for internal organs (e.g., prostate and ovaries) in the clinic has its challenges. In the first part of my talk, I will present the development of a transrectal ultrasound and photoacoustic (TRUSPA) human prostate imaging system, and its validation in various phantoms, surgically removed human prostates, in vivo mouse models of prostate cancer, all the way to the first-in-human multispectral photoacoustic human prostate imaging results. In the second part of my talk, I will introduce some new research developments in my lab. This includes results from a multimodal thermoacoustic simulation platform, novel ultrasound transducers for high throughput and wearable
photoacoustic imaging, and low-cost portable photoacoustic imaging systems.

Photoacoustic imaging relies on the generation of ultrasound waves from optically absorbing structures. The physics of photoacoustic wave generation has been compared to lightning and thunder. The interest in photoacoustic imaging has been steadily growing as optical contrast can be probed deeper in tissues compared to optical methods alone, resulting in possibly one of the most exciting new biomedical imaging techniques of the decade. Ultrasound waves produced by the absorption of light in tissue can be analyzed by methods similar to those developed to analyze ultrasound backscatter signals in the field known as ultrasound tissue characterization or quantitative ultrasound. The physics of photoacoustic wave generation can help in the interpretation of the signals detected by ultrasound transducers in photoacoustics. In the absence of exogenous optical absorbers, hemoglobin in red blood cells is the primary endogenous chromophore in tissues (as melanin is predominantly confined to the skin). The spatial distribution of red blood cells, typically confined to the vasculature, determines the frequency content of the ultrasound signals produced. Analysis of the photoacoustic signals can reveal information related to the tissue vasculature. We have applied these principles to cancer treatment monitoring and other blood pathologies. Tumor blood vessels have a distinct organizational structure compared to healthy blood vessels: typical
vessel networks are hierarchically organized, with vessels that are evenly distributed to ensure adequate oxygen and nutrient delivery. Tumor vessels are structurally different: they are torturous and typically hyperpermeable. Therapies that target the vasculature can induce changes in the vascular networks that, in principle, should be detected using photoacoustic imaging. In this presentation, we will review the techniques we have developed, which depend on the analysis of the frequency content of the ultrasound photoacoustic waves. We will show how we can use this information to filter vessels according to size with high specificity (resulting in a technique we have termed F-mode) and for non-resolvable vessels, how the frequency content of the photoacoustic signals encodes information about the size, concentration and spatial distribution of blood vessels. We also show how these techniques can be used to assess treatment response and speculate how we can use photoacoustic imaging to guide drug delivery and monitor its effects on tissues.

He is the author of the books Downtime on the Microgrid (forthcoming 2020), Ambient Commons (2013), Digital Ground (2005), and Abstracting Craft (1997) all with the MIT Press.

Whereas “the smart city” has most often been understood as a totality, top-down and always-on, in today’s climate-altered reality more agendas instead need to advance a more locally clustered, adaptive resilience. Otherwise, the most salient aspect of any future smart city is its fragility. While not a report on complexity simulations, this talk reexamines some perhaps-familiar principles of adaptive clustering from three less conventional perspectives.

John Palmer’s Bio:
John Palmer is a 1987 Chemical Engineering graduate of the University of Michigan. John worked in several roles throughout his 30 year career at Shell Oil Company, including two years as a recruiter for Shell at the University of Michigan. John began his career as a Control Systems engineer in a refinery in Houston. He then moved into team-lead positions related to control systems and electrical engineering supporting both Operations and Projects. During his career he also held a position in Human Resources, where he helped to run the internal company job-resourcing process by which engineers were allocated to their next assignment, advised engineering staff regarding career and location choices, resourced critical engineering vacancies globally, and helped establish corporate-wide engineering recruiting targets. His final assignments were as an Engineering Manager supporting major projects, which included responsibility for delivery from many different engineering disciplines (control systems, electrical, civil, structural, mechanical, materials, rotating equipment, flow assurance and process). John has worked/lived in the US, Mexico, Canada, The Netherlands and Norway, and has worked temporarily in 10 additional countries. John retired in 2018, and is excited to share his expertise with other Michigan Engineers. This event is open to all interested students in the College of Engineering.

Abstract
Recent advances in sensor arrays, imaging systems, as well as data science and AI open an exciting opportunity to rethink the way we assure and optimize performance and quality in the manufacturing industry. With the increasing availability of high-dimensional, streaming data in the industrial practice, it is now possible to predict impending anomalies and breakdowns across a manufacturing plant much earlier, and over considerably longer time horizons. Also, with the increasing availability of large time-series data, nonparametric machine learning approaches are becoming attractive for the prediction and prognosis of anomalies and breakdowns. This talk will overview three methods, each aimed at addressing a particular challenge with anomaly detection in smart manufacturing processes.
Speaker Bio
Satish T. S. Bukkapatnam received his Ph.D. and M.S. degrees in industrial and manufacturing engineering from the Pennsylvania State University. He currently serves as Rockwell International Professor with the Department of Industrial and Systems Engineering department at Texas A&M University, College Station, TX, USA. He is also the Director of Texas A&M Engineering Experimentation Station (TEES) Institute for Manufacturing Systems. His research in smart manufacturing addresses the harnessing of high-resolution nonlinear dynamic information, especially from wireless MEMS sensors, to improve the monitoring and prognostics, mainly of ultraprecision and nanomanufacturing processes and machines, and wearable sensors for cardiorespiratory processes. His research has led to over 160 peer-reviewed publications (101 published/accepted in journals and 68 in conference proceedings); five pending patents; $6 million in grants as PI/Co-PI from the National Science Foundation, the U.S. Department of Defense, and the private sector; and 14 best-paper/poster recognitions. He is a fellow of the Institute for Industrial and Systems Engineers (IISE), and the Society of Manufacturing Engineers (SME), and his work has been recognized with Oklahoma State University regents distinguished research, Halliburton outstanding college of engineering faculty, and Fulbright-Tocqueville distinguished chair awards.

Abstract
Human-robot collaboration has attracted increasing attention, both in academia and in industry. For example, in human-robot collaborative assembly, robots are often required to dynamically change their pre-planned tasks to collaborate with human operators in a shared workspace. However, the robots used today are controlled by pre-generated rigid codes that cannot support effective human-robot collaboration. In response to this need, multi-modal yet symbiotic communication and control methods have been developed. These methods include voice processing, gesture recognition, haptic interaction, and brainwave perception. Deep learning is used for classification, recognition and context awareness identification. Within this context, this seminar provides an overview of the current status of human-robot collaboration including its classification, definition and characteristics. At the end of the seminar, remaining challenges and future research directions will be highlighted.

Speaker Bio
Lihui Wang is a Chair Professor of Sustainable Manufacturing at KTH Royal Institute of Technology, Sweden. His research interests are focused on cyber-physical systems, human-robot collaboration, real-time monitoring and control, predictive maintenance, adaptive and sustainable manufacturing systems. Professor Wang is actively engaged in various professional activities. He is the Editor-in-Chief of Robotics and Computer-Integrated Manufacturing, Editor-in-Chief of International Journal of Manufacturing Research, and Editor-in-Chief of Journal of Manufacturing Systems. He has published 9 books and authored in excess of 500 scientific publications. Professor Wang is a Fellow of Canadian Academy of Engineering, CIRP, SME and ASME, the President-Elect of North American Manufacturing Research Institution of SME, and the Chairman of Swedish Production Academy.

Abstract
The technological foundations of advanced manufacturing continue to rapidly evolve as ubiquitous sensing, cloud computing and storage, and next generation controllers are introduced into the manufacturing ecosystem. This talk presents some of the technical concepts and business models that will enable new technologies and capabilities in the manufacturing sector to be rapidly deployed throughout the U.S. industrial base. Insight will be presented into next generation resilient production operations and business models that favor local and point of assembly manufacturing. The talk will conclude with a discussion of how rapidly advancing technical innovations will be propagated throughout the manufacturing enterprise, ensuring a state-of-the-art manufacturing economy. This will provide opportunities for businesses of all sizes and democratize advanced manufacturing technologies throughout the United States.

Speaker Bio
Thomas Kurfess is the Chief Manufacturing Officer for Oak Ridge National Laboratory (ORNL). In this position he is responsible for the strategic planning for advanced Manufacturing. His research focuses on the design and development of advanced systems by rapidly developing, scaling and integrating new technologies into production operations. Prior to joining ORNL, he was Professor of Mechanical Engineering and the HUSCO Ramirez Distinguished Chair in Fluid Power and Motion Control. Kurfess has also served as the Assistant Director for Advanced Manufacturing at the Office of Science and Technology Policy in the Executive Office of the President of the United States of America. In this position, he had responsibility for engaging the federal sector and the greater scientific community to identify possible areas for policy actions related to manufacturing. He earned his PhD, SM and SB in mechanical engineering, and an SM in Electrical Engineering and Computer Science from the Massachusetts Institute of Technology.
