In Vitro Platforms for the Study and Manipulation of Neutrophil Extracellular Traps

Though only recently discovered, neutrophil extracellular traps (NETs) have rapidly attracted scientific and clinical interest as a potent weapon in the arsenal of innate immunity. These structures, fibers of decondensed nuclear material on which neutrophils localize their vast antimicrobial and proinflammatory stores, are released into sites of inflammation or injury with the presumed aim of constraining and clearing bacteria. It has also been shown, however, that NETs cause substantial harm, contributing to the pathogeneses of autoimmune diseases, cancers, and thrombotic disorders as well as inciting non-specific inflammation and collateral host damage. Thus, NETs as currently understood represent a paradox in which protection seems to be outweighed by detriment. In this light, fundamental questions have arisen surrounding the identity, function, and utility of NETs in vivo. This work describes two novel platforms rationally designed to assist in understanding and contextualizing this paradox.

In the first approach, aimed at better understanding NET identity, a reductionist in vitro assay framework was iteratively developed to study NETs from the bottom up, beginning first with their DNA-histone fibrous substructure. Precise control of DNA-histone complexation yielded a robust, reproducible, and scalable structure that stood in stark contrast to low-yield and heterogeneous NET preparations. These structures, termed DNA-histone mesostructures (DHMs), mirrored both NET morphology and, to an extent, function. In doing so, DHMs provided a novel assay platform which elucidated the significant role of the isolated NET backbone in common NET-associated phenomena, such as bacterial trapping and immune activation. In addition, it permitted the confirmation and quantification of the role of the peptide LL-37 in altering NET degradation behavior. Beyond these structural studies, DHMs also yielded novel cell-based assays, including efforts to characterize the interaction between NET components and the immune system. Such studies elucidated the key role of DNA-histone synergism in NET-mediated immunostimulation, particularly amplified by the structural inclusion of non-methylated DNA. Additionally, they highlighted the importance of cell-structure proximity and contact in immune cell uptake and activation.

In the second approach, aimed at addressing the perceived pathophysiological imbalance mediated by NETs, a nanoparticulate platform was leveraged to modify cell-derived NETs in vitro with the aim of ultimately modifying them in situ. The chosen nanoparticles, hollow nanocapsules composed of polysaccharide, were internalized into neutrophils but avoided immediate NET induction; instead, they primed neutrophils for enhanced NET production only after classical stimulation. Importantly, the NETs produced by nanocapsule-loaded neutrophils were interwoven with these particles, thereby indicating significant promise for future therapeutic modification of these structures.

Though distinct in motivation and design, these two platforms demonstrate novel approaches to understanding NETs and have revealed substantial insights about both NET identity and utility as described in this work. For both, the simultaneous youth and breadth of the NET field provide a profoundly large and diverse application base. Further studies leveraging both NET-mimicking in vitro assay platforms and NET-binding nanoparticles will therefore continue to assist in the determination of both foundational and therapeutic NET biology.

As small unmanned aircraft systems (UAS) are utilized in an increasingly wide variety of commercial and civil applications, safety of flight within low altitude airspace can be improved through use of electronic geofence systems to partition the airspace. A geofence is defined as a volume of airspace with specific temporal, spatial, and permission constraints. This thesis develops geofencing as a tool for individual UAS and for managing airspace utilization through UAS Traffic Management (UTM). Permissions constraints determine which UAS may fly within each geofence. As a safety system, geofencing aims to keep the UAS within the airspace sectors (keep-in geofences) it has permission to access. Similarly, geofencing prevents the UAS from entering the airspace sectors it does not have permission to access (keep-out geofences). This thesis offers three specific contributions to geofencing.

First, a methodology is developed to enable the UTM system to build and manage the set of active geofences, ensuring a maximum of one geofence per volume of airspace at any given time. Spatial priority of geofences within the UTM system is awarded in order of request, with always active (static) geofences having top priority. Unlike static geofences, dynamic geofences appear and disappear at user-specified times and are spatially and temporally deconflicted to maximize authorized airspace volume. Polygon set operations are used to deconflict the horizontal boundaries of newly requested geofence sets from the existing UTM approved geofence set.

Second, a Triangle Weight Characterization with Adjacency (TWCA) algorithm is developed to efficiently determine whether a UAS is within a given geofence independent of the complexity of its boundary. This algorithm enables the UAS geofence module to quickly check whether the UAS is violating a geofence boundary by decomposing the horizontal boundary into triangles and tracking the occupied triangle over time through an adjacency graph. To test the performance of TWCA against the industry standard of Ray Casting, the run-time per query is calculated for randomly generated geofences and flight paths. The run-time of Ray Casting scales linearly with the number of geofence vertices while the average run-time of TWCA is constant independent of number of vertices. This time independence from geofence complexity is managed by a pre-processing step that enables real-time operation of this algorithm.

Third, to enable the UAS operator or geofence automation to intervene prior to a boundary violation, the geofence polygons are scaled to provide warning and override cues. This boundary layering algorithm utilizes a uniform and a directional buffer distance to scale keep-in geofences inward and keep-out geofences outward. The layering algorithm is designed to handle arbitrary concave polygons, with special cases identified and analyzed through Monte Carlo simulation. Multiple layering techniques are utilized in parallel to increase the likelihood of finding a scaled boundary solution. The statistical results show that the likelihood of success for inward and outward scaling decreases as buffer magnitude increases.

The contributions of this thesis are combined to form a full system simulation, from the request of a new geofence and access to an existing geofence through the prevention of the boundary violation by the UAS.

“Structural remodeling and volumetric growth in the heart”

Abstract:

Many cardiac diseases impose pressure overload leading to ventricular enlargement via the growth of cardiac myocytes and remodeling of the collagen fiber architecture. One such disease is pulmonary arterial hypertension (PAH), which primarily affects the right ventricle (RV). The effects of PAH on the functional behavior of the RV free wall (RVFW) on RV organ-level cardiac function remain largely unexplored. Computational rodent heart models (RHM) of the normal and hypertensive states can be quite valuable in gaining insights into the pathophysiology of myocardium remodeling. Recently, we developed high-fidelity biventricular finite-element RHMs using extensive data collected from rat hearts at normal and post-PAH time points. In this study, we extended our model to predict the time-course adaptations of RV under PAH from normal state to post-PAH state. Our model accounted for growth processes driven by sarcomerogenesis and collagen fibrosis, coupled with myo- and collagen fiber reorientation events. We used our model to investigate the correlations between the alterations in the wall stress, the remodeling of the RVFW microstructure, and the shape changes in the RV during the development of PAH. To the best of our knowledge, this is the first work to investigate the interaction between fiber remodeling and volumetric growth in the heart. Ultimately, the detailed description of organ-level remodeling patterns predicted by in-silico models such as ours could replace the traditional measures of RV dimensions and volume that often lead to gross and limited information on cardiac performance. Future directions of the research will also be presented.

Bicycling is an important component of future mobility for its economic, environmental, and health benefits. However, the safety issues of riding a bicycle on roadways with mixed traffic have been a major concern.

This talk covers a number of recent research projects that utilize naturalistic driving data and naturalistic cycling data to (1) examine and understand the interactions between motorists and bicyclists, and (2) support the development of automated vehicles so that they can safely interact with bicyclists on the road. The outcomes of the work could be used to support the designs of better road infrastructures, testing and benchmarking automated driving technologies, and support laws and regulations that aim to improve the safety of all road users.

Fred Feng is an assistant professor in the Department of Industrial and Manufacturing Systems Engineering at UM-Dearborn. His research interests include behavioral data analytics, human factors, cognitive ergonomics, and human-machine interaction.

ABOUT KEN WASHINGTON
Dr. Ken Washington is chief technology officer, Ford Motor Company. In this role, Washington leads Ford’s worldwide research organization, overseeing the development and implementation of the company’s technology strategy and plans, and playing a key role in the company’s expansion into emerging mobility opportunities. He reports to Jim Farley, president, New Businesses, Technology & Strategy, Ford Motor Company.

Prior to joining Ford, he was vice president of the Advanced Technology Center at Lockheed Martin Space Systems, and was responsible for leading a team of scientists and engineers in performing research and development in space science and related R&D.

Previously, he served as Lockheed Martin Corporation’s first chief privacy officer, a role in which he built the company’s privacy program, set the privacy strategy direction and established a team of privacy professionals to execute the strategy.

Washington also previously served as the vice president and chief technology officer for the Lockheed Martin internal IT organization, where he was responsible for shaping the future of the corporation’s information technology enterprise. Prior to joining Lockheed Martin in February 2007, Washington served as chief information officer for Sandia National Laboratories.

He has a bachelor’s, master’s and doctorate degree in Nuclear Engineering from Texas A&M University, is a fellow of the MIT Seminar XXI program on International Relations, and serves on the Board of McKesson Corporation.

Abstract: RNA regulation permeates neurobiology. Nociceptors are sensory neurons tasked with the detection of pain producing stimuli. Persistent changes in their activity, termed plasticity, benefit survival through injury avoidance. Nociceptors rely on cap-dependent translation to rapidly increase protein synthesis in response to pro-inflammatory signals. Comparatively little is known regarding the role of the regulatory factors bound to the 3' end of mRNA in nociceptor sensitization. Poly(A)-binding protein (PABP) stimulates translation initiation by bridging the Poly(A) tail to the eukaryotic initiation factor 4F complex associated with the mRNA cap. We have developed an RNA-based competitive inhibitor of PABP that attenuates behavioral responses to pain in mice. To identify the Poly(A) mRNAs subject to privileged translation in response to noxious cues, we have applied ribosome profiling to primary sensory neurons and tissues. A small number of transcripts are selectively translated in response to plasticity mediators. Among them is the capsid forming protein Arc. Arc has been implicated in synaptic plasticity and learning in the brain. We demonstrate that the ribosomal S6 kinase 1 is responsible for Arc production in nociceptors and describe a new role for local translation of Arc in afferent fibers. Collectively, our findings uncover mechanisms and targets of RNA control in sensory neurons that can be exploited to disrupt pain signaling.

REQUIREMENTS:
To qualify for the Accelerator, you must be:
Graduating with a Computer Science or related Bachelor's degree.

APPLICATION PROCESS:
To apply for the Atomic Accelerator, you'll need to participate in the Atomic Games (October 25 & 28). The Games are a programming competition for college students pursuing a bachelor’s degree in computer science, computer engineering, or a related program.

For the Atomic Games, we’ve created an exciting turn-based game. We will provide the game and an artificial intelligence implementation for participants to play against. The game will coordinate moves between two AI engines, invoking an engine with the current game state and expecting the engine to return its next move in a reasonable amount of time. Participants will create an artificial intelligence algorithm, then face off against one another for the chance to win $500. During the Games, Atomic will be on the lookout for people to join our team in 2020 as full-time developers.

Challenges in Prosthetic Limbs: Design, Control, Use, and Utility
Presented by: Assistant Professor Peter G. Adamczyk of the University of Wisconsin-Madison

Abstract
Artificial limbs offer an opportunity to improve movement through biomimetic devices. One approach is to directly replace joint function, but achieving humanlike performance is challenging for design, control, cost and longevity of the systems. An alternative approach is to exploit biomechanical workarounds for lost function rather than directly replacing it. This presentation will describe several such “semi-active” prostheses – low-power systems that modulate their mechanical properties but cannot power body movement – that aim to add adaptability and versatility with minimal addition of weight, height, complexity, power demand and cost.
Another challenge in rehabilitation and assistive technology is determining which among several interventions is most beneficial to everyday movement. “Real-world” assessment using wearable sensors is a popular approach, but current analysis techniques struggle to reduce days-long data sets to generalizable knowledge. The second part of this presentation will describe this challenge and a novel approach to data reduction aimed at enabling lab-like scientific findings from long-term wearable data sets, with upcoming application to prosthetic ankle-foot systems.

Bio
Dr. Peter Adamczyk earned degrees in Mechanical Engineering from Case Western Reserve University (B.S.) and the University of Michigan (M.S. and Ph.D) in the areas of Robotics and Biomechanics. He spent several years running a startup company dedicated to advancing the science and technology of lower-limb prosthetics and real-world motion assessment. He is now an Assistant Professor at the University of Wisconsin–Madison where he directs the Biomechatronics, Assistive Devices, Gait Engineering and Rehabilitation Laboratory (UW BADGER Lab, http://uwbadgerlab.engr.wisc.edu).

Dr. Adamczyk’s research aims to enhance physical and functional recovery from impairments affecting walking, running, and standing. Core foci include the design of semi-active foot prostheses for gait restoration after amputation; wearable sensors for movement assessment during real-life activities; and rehabilitation robotics to explore motor learning and neural adaptation.

Neural Interfacing with Dorsal Root Ganglia: Anatomical Characterization and Electrophysiological Recordings with Novel Electrode Arrays

Dorsal root ganglia (DRG), the hubs of neurons conducting sensory information into the spinal cord, are promising targets for clinical and investigative neural interface technologies. DRG stimulation is currently a tertiary therapy for chronic pain patients, which has an estimated prevalence of up to 40% of adults in the United States. In pre-clinical studies, combined neural recording and stimulation at DRG has been used as part of novel closed-loop systems to drive activity of the limbs or the urinary system. This suggests a potential role for clinical DRG interfaces to assist, among other patient groups, the nearly 300,000 spinal cord injured patients in the United States.

To maximize the utility of DRG interfaces, however, there remains a strong need to improve our understanding of DRG structure. Neural interface technologies for both stimulation and recording rely heavily on the spatial organization of their neural targets. To record high-fidelity neural signals, a microelectrode must be placed within about 200 µm of a neural cell body. Likewise, effective neural stimulation is believed to act on a subset of DRG axons based on their size and location. The spatial organization of DRG, however, has not been previously quantified. In this thesis, I demonstrate a novel algorithm to transform histological cross-sections of DRG to a normalized circular region for quantifying trends across many samples. Using this algorithm on 26 lumbosacral DRG from felines, a common preclinical DRG model, I found that the highest density of neural cell bodies was in the outer 24% radially, primarily at the dorsal aspect. I extended this analysis to a semi-automated cross-DRG analysis in 33 lower lumbar DRG from 10 human donors. I found that the organization of human DRG was similar to felines, with the highest density of cell bodies found in the outer 20-25% of the DRG, depending on spinal level. I also found a trend toward lower small-axon density at the dorsal aspect of L5 DRG, a key region for stimulation applications.

To take advantage of this quantitative knowledge of DRG organization, future neural interfaces with DRG will require more advanced technologies. Standard silicon-based electrode arrays, while useful for short-term DRG recordings, ultimately fail in chronic use after several weeks as a result of mechanical mismatch with neural tissue. In this thesis, I demonstrate sensory recording from the surface and interior of sacral DRG during acute surgeries using a variety of flexible polyimide microelectrode arrays 4 μm thick with minimum site separation of 25 to 40 μm. Using these arrays, I recorded from bladder and somatic afferents with high fidelity. The high density of sites allowed for neural source localization from surface recordings to depths 25 to 107 µm. This finding supports the anatomical analysis suggesting a high density of cell bodies in the dorsal surface region where the surface array was applied. The high site density also allowed for the use of advanced signal processing to decrease analysis time and track neural sources during movement of the array which may occur during long-term behavioral experiments.

This thesis represents significant advances in our understanding of DRG and how to interface with them, particularly related to the ways anatomy can inform development of future technologies. Going forward, it will be important to expand the anatomical maps based on organ function and to test the novel flexible arrays in chronic implant experiments.

“Multi-Omics Systems Biology Approach to
Vascular Biology”

Abstract:

Over the past decade, we have investigated the mechanisms associated with shear stress response in endothelial cells using systems biology approaches [e.g. 1-8]. In the most recent granting period, we have carried out longitudinal analysis of distinctive shear stress response to pulsatile and oscillatory stress corresponding to normal and disturbed flow in blood vessels. Our studies have provided fundamental insights into temporally regulated mechanisms as well as a systems-level view of stress response. Our key findings also included implications of long noncoding RNA molecules – LEENE and LINC341 - which appear to have dramatic consequences on endothelial homeostasis. The regulation by LINC RNAs entails the coupling between KLF4 and genes involved in endothelial function. In addition, we explored the interplay between endothelial cells and macrophages in atherogenesis. These studies reveal a novel mechanisms in which the transcription factor KLF4 regulates the enzyme cholesterol-25-hydroxylase and the nuclear hormone receptor Liver X Receptor. Our key findings indicate that KLF4 transactivates cholesterol-25-hydroxylase and liver X receptor, thereby promoting the synergistic effects between ECs and macrophages to protect against atherosclerosis susceptibility.

This workshop will provide an overview of several different types of interviews and how to best prepare for each, including behavioral, technical, case, and phone/Skype interviews. We will review strategies for answering interview questions, such as the STAR format, and discuss what to emphasize when answering interview questions. Preparing for common interview questions is only one part of the process - learn what to wear and bring to an interview and how to follow up with an employer after the interview. Come learn how to ace the interview!

There is an increasing body of literature on understanding the process of evacuation, and it focuses both on people's behavior and emergency management. We need to consider aspects of interaction among people under nonotice disaster for planning with evacuation demand. This seminar examines the effect of social interaction on discrete choice during the network formation process. The primary objective is to evaluate influence on risk regarding from others in greater detail while considering how influencers and network structures affect one-to-one interactions. This talk examines an analytical framework for simultaneous evaluation of social interaction and social network formation. The framework is well suited to dynamic disaster situations because local interaction heavily influences human decision-making, and because network formation changes over time.

Junji Urata is an assistant professor in Department of Civil Engineering at University of Tokyo. His research interests include behavioral modeling, analytics of traffic demand, social interaction, dynamic programming, and high performance computing.

The demand for lightweighting of our infrastructure and transportation equipment has seen a significant growth the last two decades; this need is expected to increase as reduction of carbon footprint and energy consumption will be major drivers in the decades ahead. Aluminum has played a pivotal role in this lightweighting movement and will continue to do so. There is a need for high performance Al alloys such as enhanced modulus, usage at elevated temperatures, and damage resistant alloys, etc. In this presentation a retrospective and a prospective view of alloy design for Al will be reviewed and discussed.

Diran Apelian is the Alcoa-Howmet Professor of Engineering and Founding Director of the Metal Processing Institute (MPI) at Worcester Polytechnic Institute (WPI). He is also Distinguished Professor of Materials Science and Engineering at the University of California, Irvine.
He received his B.S. degree in metallurgical engineering from Drexel University in 1968 and his doctorate in materials science and engineering from MIT in 1972. He worked at Bethlehem Steel’s Homer Research Laboratories before joining Drexel University’s faculty in 1976. At Drexel he held various positions, including professor, head of the Department of Materials Engineering, associate dean of the College of Engineering and vice-provost of the University. He joined WPI in July 1990 as WPI’s Provost. In 1996 he returned to the faculty and led the activities of the Metal Processing Institute, which he founded. In 2019, he joined the faculty at UCI.
He is credited with pioneering work in various areas of metal processing – specifically in molten metal processing, alloy development, solidification and casting, surface engineering, spray casting/forming, and powder metallurgy. During the last decade, he has worked on sustainable development issues, and particularly, resource recovery, reuse, and recycling. Apelian is the recipient of many distinguished honors and awards – national and international; he has over 700 publications to his credit; and serves on several technical, corporate and editorial boards. During 2008/2009, he served as President of TMS. He served as Chair of the ASM Educational Foundation Board of Trustees (2016-2018). Apelian is a Fellow of TMS, ASM, and APMI; he is a member of the National Academy of Engineering (NAE), National Academy of Inventors (NAI), and the Armenian Academy of Sciences (AAS). He is a recipient of the 2016 Bernard Gordon Prize for Innovation in Engineering Education (with Kris Wobbe, Art Heinricher and Rick Vaz).

Abstract
Multi-component flows play a central role in engineering problems ranging from the design of biomedical devices for therapy to naval and aircraft development. I will consider cellular, cavitating, and droplet flows as canonical examples of such flows. Each case is treated using novel computational techniques for simulation. This includes a sub-grid disperse flow model that is based upon particle population moments and improved via recurrent neural networks. The high-fidelity spectral and discretely-conservative interface-capturing methods used to solve for the flow will be discussed, including a presentation of our new open-source solver, MFC, that implements them. These large-scale simulations are complemented by novel analyses based-upon non-modal stability theory, chaotic dynamical systems, and stochastic and data-driven techniques. These are interpreted as they apply to microfluidics, rheometry, and even feeding humpback whales.

Bio
Dr. Spencer Bryngelson is a Senior Postdoctoral Scholar at the California Institute of Technology, working with Professor Tim Colonius. Previously, he was a Postdoctoral Researcher at the Center for Exascale Simulation of Plasma-Coupled Combustion (XPACC), a PSAAP II center. He received his PhD and MS in Theoretical and Applied Mechanics from the University of Illinois at Urbana–Champaign in 2017 and 2015, respectively, working with Professor Jonathan Freund. In 2013, he obtained BS degrees in both Mechanical Engineering and Engineering Mathematics from the University of Michigan–Dearborn. His research lives at the intersection of fluid dynamics and computational physics, with a focus on biomedical, defense, and environmental applications. In pursuit of this, he develops high-performance software, physical models, numerical methods, and techniques for physics-based and data-driven analysis

“Accelerating the computational discovery of catalyst design 
rules and exceptions with machine learning ”

ABSTRACT

Over the past decade, first-principles computation has emerged as a powerful complement to experiment in the discovery of new catalysts and materials. In many cases, computation has excelled most in distilling rules for catalyst structure-property relationships in well defined spaces such as bulk metals into descriptors or linear free energy relationships. More development is needed of computational tools for them to show the same promise in emerging catalytic materials such as single-site metal-organic framework catalysts or single atom catalysts that have increased promise of atom economy and selectivity. In this talk, I will outline our efforts to accelerate first-principles (i.e., with density functional theory, or DFT) screening of open-shell transition metal catalysts with a focus on challenging reactions (e.g., selective partial hydrocarbon oxidation). We have developed tools that not only automate simulation but can be autonomously driven by decision engines that predict which simulations are most promising to be carried out. We also develop neural network machine learning models to accelerate prediction of catalyst reaction energetics and properties at a fraction of the cost of DFT. Paired with new estimates of when such models are reliable, I will show how we rapidly evaluate properties of 10k-100k catalysts in a fraction of the time that conventional first-principles simulation would require. We use such tools to accelerate the identification of design rules and exceptions to expectations when applied to the wider space of emerging single-atom and single-site catalysts.

SHORT BIO

Professor Heather J. Kulik is an Associate Professor in the Department of Chemical Engineering at MIT. She received her B.E. in Chemical Engineering from the Cooper Union for the Advancement of Science and Art in 2004 and her Ph.D. from the Department of Materials Science and Engineering at MIT in 2009. She completed postdoctoral training at Lawrence Livermore and Stanford, prior to joining MIT as a faculty member in November 2013. Her research in accelerating computational modeling in inorganic chemistry and catalysis has been recognized by a Burroughs Wellcome Fund Career Award at the Scientific Interface, Office of Naval Research Young Investigator Award, DARPA Young Faculty Award, NSF CAREER Award, the AAAS Marion Milligan Mason Award, and the Journal of Physical Chemistry Lectureship, among other awards.

Abstract
Multi-component flows play a central role in engineering problems ranging from the design of biomedical devices for therapy to naval and aircraft development. I will consider cellular, cavitating, and droplet flows as canonical examples of such flows. Each case is treated using novel computational techniques for simulation. This includes a sub-grid disperse flow model that is based upon particle population moments and improved via recurrent neural networks. The high-fidelity spectral and discretely-conservative interface-capturing methods used to solve for the flow will be discussed, including a presentation of our new open-source solver, MFC, that implements them. These large-scale simulations are complemented by novel analyses based-upon non-modal stability theory, chaotic dynamical systems, and stochastic and data-driven techniques. These are interpreted as they apply to microfluidics, rheometry, and even feeding humpback whales.

Bio
Dr. Spencer Bryngelson is a Senior Postdoctoral Scholar at the California Institute of Technology, working with Professor Tim Colonius. Previously, he was a Postdoctoral Researcher at the Center for Exascale Simulation of Plasma-Coupled Combustion (XPACC), a PSAAP II center. He received his PhD and MS in Theoretical and Applied Mechanics from the University of Illinois at Urbana–Champaign in 2017 and 2015, respectively, working with Professor Jonathan Freund. In 2013, he obtained BS degrees in both Mechanical Engineering and Engineering Mathematics from the University of Michigan–Dearborn. His research lives at the intersection of fluid dynamics and computational physics, with a focus on biomedical, defense, and environmental applications. In pursuit of this, he develops high-performance software, physical models, numerical methods, and techniques for physics-based and data-driven analysis

Abstract:
A dense plasma focus (DPF) is a relatively compact coaxial plasma gun which completes its discharge as a Z-pinch. These devices have been designed to operate at a variety of scales in to produce short (<100 ns) pulses of ions, X-rays, or neutrons. LLNL has recently constructed and brought into operation a new device, the MJOLNIR (MegaJouLe Neutron Imaging Radiography) DPF which is designed for radiography and high yield operations. This device has been commissioned over the last year and has achieved neutron yields up to 9x10^11 neutrons/pulse at 2.2 MA pinch current while operating at up to 1 MJ of stored energy. MJOLNIR is equipped with a wide range of diagnostics, including activation foils, neutron time of flight detectors, a fast framing camera, optical light gates, and a time-gated neutron and x-ray imager. LLNL also runs unique particle-in-cell (PIC) simulations of DPF in the Chicago code, and has been able to gain significant insight into the physical factors that influence neutron yield. To that end, MJOLNIR is one of the first DPFs whose design and continual upgrades are heavily influenced by predictive modeling. In this presentation, we will describe insights from modeling, device operation, and recent results. Preliminary x-ray and neutron images will also be presented.

About the Speaker:
Dr. Andrea Schmidt is group leader of the Plasma Engineering Group and Associate Program Lead for Pulsed Power Fusion Plasmas at Lawrence Livermore National Laboratory (LLNL). She received her Ph.D. in Physics from MIT and her BS in Physics from the U. of California/Berkeley. She joined LLNL as a postdoctoral researcher in 2011 and joined the staff in 2013. As a postdoc, Schmidt was involved in electrical grid research, and modeling the dense plasma focus (DPF) device. She performed the first kinetic modeling of a DPF, demonstrating that a particle approach was needed to capture beam formation and neutron yield. She is now leading several projects in DPF research including the development of a large MJ-class DPF experiment built for flash neutron radiography. Schmidt also led modeling and experimental efforts for magnetron sputtering and was part of a team investigating a shear-flow-stabilized z-pinch configuration for controlled fusion.

In the last decade, the rapid development of communication technology, coupled with artificial intelligence with big data, had significantly impacted and will continue to transform people’s daily travel. This creates significant opportunities for innovation in the field of transportation engineering. Such opportunities are manifested by the availability of massive mobility service data that is not seen before and the rapid development of autonomous vehicle technologies. In this talk, Dr. Liu will discuss the future trends for transportation engineering, drawing his experience as a seasoned transportation researcher and in the last two years as a business executive in a mobility service company. He will also discuss his research work on connected vehicle based traffic signal control and the process of transferring the research results into a commercial product, which is currently being offered as software as a service on the market. It is his hope that this talk can shed some light on opportunities and challenges for future transportation research and education.

A number of hot topics in aviation right now — such as small UAS, urban air mobility, eVTOL aircraft — depend on new multirotor aircraft configurations. We know that these vehicles can fly in idealized conditions, but will it take for their use to really “take-off” in society? This presentation will start by reviewing the enablers of these new vehicles. Then, we’ll note a number of pressing issues being examined at Penn State that may limit their wide-spread use. For example, small rotors can easily accrete ice in less-than-perfect atmospheric conditions. When designed for maximum performance, they can be too noisy to operate in an urban environment — requiring trade-offs between vehicle performance and societal acceptance, and meriting a careful engineering design of their landing and take-off procedures.

These vehicles typically depend on new methods of flight control and the business cases for many of them assume highly automated operations allowing for reduced human staffing levels. In a broader sense, these vehicles, and their operations, are so novel compared to traditional methods for certification and operational approval that the aviation community needs to envision new bases for safety cases, including new concepts for human-autonomy teaming, new requirements for adaptive, contextualized behavior, and new methods for modeling and assessing safety.

About the Speaker...

AMY R. PRITCHETT is a professor and head of the Department of Aerospace Engineering at the Pennsylvania State University. Previously, Dr. Pritchett was on the faculty of the Schools of Aerospace Engineering and of Industrial and Systems Engineering at the Georgia Institute of Technology from 1997 to 2017, and she also served via an IPA as the Director of NASA’s Aviation Safety Program in 2008 and 2009. Her research focuses on the intersection of technology, expert human performance and aerospace operations, with a particular focus on designing to support safety. She has received the AIAA Lawrence Sperry Award, the RTCA William Jackson Award and, as a member of the Executive Committee of the Commercial Aviation Safety Team, the 2008 Collier Trophy. She chaired the National Research Council Committee examining FAA Air Traffic Controller Staffing Levels and Implications for FAA Budget, and for Implementation of the Next Generation Air Transportation System, and recently served as a member on other NRC committees reviewing the FAA Certification Research Plan, Methods for Assessing the Safety of UAS Operations in the NAS, and NASA’s astronaut corps. Teaching HFE To BME Undergraduate and Graduate Students: Lessons & Tips

Abstract:
Dr. Gosbee will present some short examples of how he teaches the basics of human factors engineering (HFE) to the biomedical engineering (BME) learners. For instance, one interactive group exercise is applying the Stroop Effect to design of labeling for medical gas cylinders. He will show how usability testing and HFE aspects of risk management fit into BME senior and graduate design courses (as co-instructor). The focus is on methods and structure required by FDA and international standards (e.g., IEC 62366). Additionally, a short overview of how he also injects some HFE into other BME courses like “Needs Finding” (BME 499, Dr. Rachael Schmedlen, lead) and Regulatory science (BME 588, Dr Joan Greve, lead) will be presented.

Bio:
John Gosbee, MD, MS teaches human factors engineering (HFE) and patient safety at the University of Michigan Departments of Internal Medicine and Biomedical Engineering. He leads development of patient safety curriculum for several medical and surgical residencies. He teaches HFE for several BME design courses, including capstone design courses, observation course, and regulatory science. He also provides HFE consultation to UM research projects that involve new device design (e.g., teleophthalmology). He has been visiting professor at dozens of universities, including Penn, Johns Hopkins, and Yale University. He has received two national awards for patient safety design (ISMP’s “Cheers Award” and AAMI’s “Career Achievement Award”. Among dozens of other publications, he edited and co-wrote the book, Using Human Factors Engineering to Improve Patient Safety. Previously, Dr Gosbee worked at Department of Veterans Affairs - National Center for Patient Safety, Assistant Professor at Michigan State University, and NASA on development of space-based medical facilities.

"Precision biomaterial platforms to probe valvular fibroblast activation"

ABSTRACT

Aortic valve stenosis (AVS) is a progressive disease characterized by aberrant stiffening of the aortic valve, leading to inadequate blood flow from the left ventricle and eventual heart failure. AVS is currently treated exclusively with valve replacement surgeries, which may be avoided if effective small molecule therapeutics could be identified to slow or reverse AVS progression. During AVS progression, a large fraction of valvular interstitial cells (VICs) differentiate into pathogenically activated myofibroblasts, which contribute to excessive matrix deposition and eventual valve leaflet stiffening. Effective small molecule therapeutics intended to reverse myofibroblast activation remain elusive, owing to the inherent heterogeneity of the cellular microenvironment from patient to patient. For example, male patients show increased calcification and female patients show increased tissue fibrosis in the aortic valve microenvironment, leading to differential drug responses. Considering these clinical observations, we seek to engineer precision biomaterial microenvironments to recapitulate sex- and patient-specific AVS progression in vitro and identify molecular mechanisms that mediate reversal of myofibroblast activation. Precision biomaterials are defined here as engineered environments that enable the evaluation of how sex- and/or patient-specific variables may influence disease progression. As a strategy to evaluate molecular mechanisms guiding AVS progression, we utilize poly(ethylene glycol) (PEG) hydrogels as precision in vitro platforms to probe various biochemical and mechanical cues that activate male and female VICs to a myofibroblast state, as well as cues that reverse activation (Fig. 1A). My talk will focus on two vignettes where PEG hydrogels were used to evaluate how male and female VICs activate to and reverse from a myofibroblast state in response to different cues, including (1) serum factors from AVS patients before and after a valve replacement procedure and (2) small molecule inhibitors of myofibroblast activation. Collectively, our work demonstrates how engineered hydrogel matrices enable an increased understanding of the molecular mechanisms guiding myofibroblast activation and reversal, which may provide a critical bridge toward sex- and patient-specific small molecule AVS therapies.

SHORT BIO

Dr. Brian Aguado is currently an NIH K99 and Burroughs Wellcome Fund postdoctoral fellow at the University of Colorado. His current research is focused on developing precision biomaterials for applications in personalized medicine. Dr. Aguado completed his MS and PhD in biomedical engineering from Northwestern University and his BS degree in biomechanical engineering from Stanford University. He also obtained his certificate in Management for Scientists and Engineers from the Kellogg School of Management at Northwestern. Dr. Aguado is also a dedicated science communicator outside of the lab and seeks to engage underrepresented populations in the sciences. He is the former president of the Postdoctoral Association of Colorado, organized seminars featuring underrepresented thought leaders in STEM for CU Café, and served on the executive board for Project Bridge Colorado. Most recently, he co-founded LatinXinBME, a new social media initiative dedicated to building a diverse and inclusive community of Latinx biomedical engineers and scientists to support each other personally and professionally through their careers.

Speaker: Amy Marconnet - Associate Professor at Purdue University

Abstract
Nanostructuring material s allows independent control of multiple material s properties. High conductivity material s such as carbon nanotube forests are useful as thermal interface materials (TIMs) for dissipating power in electronic devices, while low conductivity material s like nanoporous silicon for thermal barrier coatings and enhanced thermoelectric performance. Beyond thermal transport, storage of thermal energy is critical for effective heat removal for applications involving
highly-transient heat fluxes, and during material processing. Often to achieve the desired functionality, multiples material s are combined together to form heterogeneous composites. For example, in lithium-ion batteries, the particulate active material s (with micro- and nano-scale features) are sandwiched between metal electrodes and polymer-based separators with microscale thicknesses to form macroscale battery cells. This seminar will discuss methods to understand and
control thermal transport and development of accurate and reliable experimental and analytical techniques for thermal characterization across multiple length scales. Further, I will highlight the integration of material synthesis with thermal property measurements and physics-based analysis to provide new avenues for improved material s and device performance.

Bio
Amy Marconnet i s an Associate Professor of Mechanical Engineering at Purdue University. She received a B.S. in Mechanical Engineering from the University of Wi Wisconsin – Madison in 2007, and an M.S. and a PhD in Mechanical Engineering at Stanford University in 2009 and 2012, respectively. She then worked briefly as a postdoctoral associate at the Massachusetts Institute of Technology before joining the faculty at Purdue University in 2013. Research in the Marconnet Thermal and
Energy Conversion (MTEC) Lab integrates metrology and analysis of underlying transport mechanisms with design and development of nanostructured material s for heat transfer and energy conversion applications.

**Fall 2019 KICK-OFF WORKSHOP SEPTEMBER 23RD**

Sandwiches and Science: Training (for) Better Presentations marks the third run of the professional development event hosted by Tau Beta Pi aimed at providing Michigan Engineering graduate students the opportunity to enhance their scientific communication skills. The series will be co-hosted/sponsored by TBP and the graduate societies of MSE, ECE, ChE, and MACRO and also sponsored by the Office of Student Affairs! As "learning-by-practice" event, it aims to help students learn how to effectively convey the "big picture" value of their research to a diverse audience, while also engaging a dialog of science and engineering research among graduate students across the entire College of Engineering. The event is aimed primarily at graduate students planning to take their candidacy exam, but anyone is welcome to participate! We will host 7-10 events each term, and event dates/times will be announced on a rolling basis.

Each session is structured to have student speakers (2-3 per session) make a timed (15-20 min) presentation on their graduate research to a broad engineering audience and a communications expert panel (3-4 panelists). Our expert panelists will provide constructive feedback to the speakers (and the audience), highlighting the positive aspects of each presentation and also indicating opportunities for improvement. This structure will allow for the speakers to receive specific feedback on their communication skills, while also providing the audience with generalized guidelines for good scientific communication.

TITLE: Advancing Evidence-based Biomedical Engineering Education in Real Time

Undergraduate biomedical engineering (BME) programs typically consist of courses from several different academic departments combined with BME-specific courses taught by faculty trained in a variety of disciplines. While some students embrace this diversity in courses and disciplinary perspectives, many struggle with how to translate these experiences into career opportunities. BME students are often concerned that they are perceived as a “jack of all trades, masters of none.” In 2016, the BME Department sought to find new ways to integrate BME professional practice into the curriculum.

Informed by organizational change theory, we asked: 1) Is there potential for change; 2) what strategies facilitate change; and 3) how can these strategies be implemented? As a result, we developed an Instructional Design Sequence, a new approach to instruction in which students, post docs, and faculty create short modules that use evidence-based teaching practices to expose BME students to BME professional practice.

This presentation describes how the Instructional Design Sequence was conceived and demonstrates how theory can be used to inform practice. The resultant Sequence is a transferrable model for transforming engineering education, offering a mechanism for integrating new career-relevant curriculum into undergraduate curriculum, while training future educators in evidence-based instructional practices.

Biography
Tony S. Hsu is the grandson of Xu Zhimo. He was born in Shanghai shortly after the end of World War II. As a toddler, Hsu and his sisters were raised by his grandmother, Zhang Youyi, while his parents pursued their studies in America.

In the late 1940s, Zhang and her young charges left China amidst national political turmoil and settled in Hong Kong. At age six, Hsu and his sisters emigrated to New York to join their parents and begin a new life in America. Hsu ultimately received his bachelor’s in electrical engineering from the University of Michigan and doctorate in applied physics from Yale University. He has been an executive for several technology companies. Hsu lives with his fashion designer wife, Lily Pao Hsu, and his filmmaker daughter, Alexandra, in Southern California. Chasing the Modern is his first book.

Impacting Healthcare Costs with a University of Michigan innovation – FlexDex

Dr. Kent Bowden, General Surgery, Cadillac Hospital, Munson Healthcare System

Abstract
Driven by minimally invasive procedures that require intensive suturing, the adoption of robotics in general surgery has accelerated dramatically in spite of high costs and steep learning curve. While big hospitals are able to absorb the cost of owning and maintaining multi-million dollar DaVinci surgical robots and associated infrastructure, small community hospitals face a difficult decision.

In this talk, I will present this dilemma faced by Cadillac Hospital, which serves the rural community of about 80,000 residents in seven counties in northern Michigan. A few years ago, we had to decide to either invest significant time, money, and personnel in creating and running a robotics program, or lose our patients and surgeons to bigger hospitals and medical centers. This would be a loss for the
local community, economy, and quality of living. Instead, we looked for alternate options. There weren’t many but the most promising one seemed to be a technology from our own state – FlexDex – created at the University of Michigan.

I will describe how I ended up adopting FlexDex and pioneering complex surgical procedures with it just as they would be performed on the DaVinci surgical robot but at much lower costs to the hospital. I have now performed over four hundred FlexDex procedures including hernia repair, colon resection, fundoplication, and hysterectomy. The savings from this decision and the “notoriety” that this brought us has expanded our surgical efficiency as well as capabilities.

FlexDex is now spreading all over the world and is being adopted in urology, gynecology, cardiothoracic, bariatric, and whipple procedures among others. I will discuss the future potential of this technology in providing greater access to minimally invasive surgery world-wide at an affordable cost. I will also discuss ongoing technology needs and challenges that remain in surgery that might
inspire new research and innovation at universities.

Dr. Kent Bowden, D.O. is a General Surgeon at Cadillac Hospital, Munson Healthcare. He trained at the College of Osteopathic Medicine, Michigan State University and graduated in 2005. He completed his Residency at Ingham Regional Medical Center in Lansing, MI in 2010, and joined private practice in a rural hospital where he could have a broad-spectrum practice. His objective as a surgeon is to
provide cost-effective, personalized and world-class care to patients in his local community. In addition to pioneering cases with FlexDex, he enjoys coaching basketball at his children's school and spending time outdoors with his family. Dr. Bowden is a Fellow of the American College of Osteopathic Surgeons (FACOS).

Join the Michigan Engineering community for a conversation with leaders from the nuclear industry, including former leadership from the Department of Energy. Dr. Monica Regalbuto, Idaho National Laboratory, served as former Assistant Secretary for Energy and Environmental Management for the U.S. Department of Energy where she was responsible for clean-up of sites, including Rocky Flats and the Hanford site. River Bennett is a Nuclear Policy Analyst for Nuclear Innovation Alliance which supports entrepreneurialism and commercialization of advanced nuclear energy systems with the goal of providing more economically competitive, carbon-free energy to the world. He is an advocate with Students for Nuclear. Dr. Sola Talabi is a consultant with Pittsburgh Technical, providing expertise in risk management consulting within the nuclear energy industry. The panel will be moderated by Dr. Ann Verhey-Henke, Strategic Director of the U-M Center for Socially Engaged Design.

Panelists will provide insight in several areas, such as:
-What is the status of clean-up of sites like Rocky Flats and Hanford? What big challenges are remaining?
-What are the global implications of nuclear energy growth? Where is nuclear energy growing most significantly? Does this represent a challenge or a new opportunity?
-Is nuclear energy safe? Can it be contained? How do we assess risk in this area?
-How accurate is the portrayal of Rocky Flats as presented in “Full Body Burden” by Kristen Iversen?

Dr. Gui started his career in 1969 and has served as contractor chief scheduler, program manager, construction manager, forensic scheduler, and expert witness. He has pioneered innovations in project management throughout his 50-year career. He holds four US patents and has numerous patents pending on his graphical path method. Dr. Gui pursued MS studies at Vanderbilt University in 1966-1967 and obtained his PhD in civil engineering from Michigan in 1972. From 1973-1989, Dr. Gui taught two graduate courses in network-based project scheduling in the College of Engineering, Civil & Environmental Engineering, at Michigan.

Abstract:
NASA is preparing to demonstrate high-power electric propulsion on the Power and Propulsion Element (PPE), the first element of a human-operated Gateway station that will orbit the Moon. The Hall thrusters (HTs) being flown on the PPE will operate at three times the power of current state-of-the-art HTs and utilize magnetic shielding, which greatly increase the wear life of the thrusters. Associated with these advances in technical capability are uncertainties regarding the plasma properties of the exhaust plume, particularly the low density plume exiting sideways from the thrusters. To better predict the erosive power of the plasma plume and how the plume might interact with the PPE and future spacecraft, NASA is flying the Plasma Diagnostics Package (PDP). The PDP data will refine plasma models for predicting the behavior of high-power HT systems on future missions. This presentation will describe a brief history of the PDP project, basic Hall thruster plume physics, the plasma physics behind the PDP sensor design, and current status of the PDP project.

About the Speaker:
Wensheng Huang received a PhD in Aerospace Engineering from the University of Michigan in 2011 and BS degrees in Mechanical and Nuclear Engineering from the University of California, Berkeley in 2006. His dissertation was on the use of optical diagnostics to study Hall thruster erosion under the tutelage of Prof. Alec D. Gallimore. Wensheng is currently a researcher in the Electric Propulsion Systems branch at the NASA Glenn Research Center. He is the Principle Investigator for the Plasma Diagnostics Package (PDP) that will be flying on the Power and Propulsion Element (PPE), the first element of NASA's Moon-orbiting Gateway. He is also the diagnostics lead for the Solar Electric Propulsion Hall thruster project, which will also be flying on the PPE.

“Radically Simplifying hMSC Manufacturing Scale-up and Regenerative Medicine Product Development (and Building a Business around Regenerative Medicine Manufacturing Technology)”



Abstract:

Human Mesenchymal Stem/Stromal Cells (hMSCs) and extracellular vesicles (hMSC-EVs) are leading platforms for development of regenerative medicine therapies. Multiple bottlenecks exist during discovery, product development, manufacturing process development, and cGMP manufacturing leading to timelines from discovery through first in man testing often averaging 7-11 years. RoosterBio has developed and commercialized novel hMSC bioprocess systems that are designed for rapid process development and cGMP tech transfer to radically reduce development timelines to as little as 2-4 years. Requirements for implementation of scalable bioreactor production systems will also be addressed, and data will be presented from 50L scale microcarrier-based processes with CQAs for both hMSCs and EVs.
