"A Double-Edged Sword”: Genetic Data Sharing and Implications for the Learning Health System

In contrast to the laborious and expensive process of generating genetic datasets de novo, academic genetic researchers are increasingly using large and inexpensive “secondary” research datasets held by government, consortia, and industry for their work. Choosing between different kinds of data providers is about more than just convenience, however, it can also have important implications for the kind of science advanced and to which communities it will generalize. This talk will explore the factors driving researchers to select certain datasets for their work as well as their experiences sharing to, as well as using, shared data resources. As researchers wait for the new National Institutes of Health’s “Policy for Data Management and Sharing” to go into effect in January 2023, this talk will explore who ultimately carries the burden of increasing data sharing requirements

To study the neural circuitry, the action of cells under the context of others, one would precisely measure and perturb specific neuronal populations and molecules in behaving animals who are specifically engaged in performing the computation or function of interest. The data set of millions of neurons firing together underlying a behavior are required to develop and refine theories (hypotheses) explaining animal behavior in terms of brain physiology. The focus of my lab is to develop novel genetically encoded indicators based on fluorescence proteins, especially focusing on direct and specific measurement of myriad input signals with needed spatial and temporal resolutions. In this talk, I will discuss our recent progress in developing and applying a new suite of genetically encoded indicators of neural activity. I will discuss the design, characterization, and applications of these genetically encoded indicators for both in vivo imaging and drug discovery. In combination with calcium imaging and optogenetics, the sensors are well poised to permit direct functional analysis of how the spatiotemporal coding of neural input signaling mediates the plasticity and function of target circuits.

The Tian Laboratory for Optical Neurophysiology engineers biosensors and optical probes for monitoring and controlling brain activity in living, behaving research subjects. When combined with behavioral and circuit manipulations, these tools can reveal the brain mechanisms underlying the control of various behaviors in health and disease and serve as drug discovery platforms for the identification of novel therapeutic targets. Abstract: The human oral microbiome typically contains over 700 different microbial species. How they interact can shape the microenvironment throughout the human body, as these interactions are paramount to maintaining oral and overall systemic health. Previous epidemiological studies have linked dysbiosis of oral microbes with several chronic ailments such as cardiovascular disease, poor glycemic control in diabetics, and rheumatoid arthritis. Recent advances in technology, including next-generation sequencing (NGS), have revealed the complexities of the oral microbiome, but the role of microbial and molecular interactions during systemic disease progression remains poorly understood. Here we show significant shifts in the oral metabolite profile during the progression of early-stage periodontal disease (gingivitis) and identify several key metabolites as quantitative measures for gingivitis progression. In addition, our study reveals a previously unknown metabolic lag in recovery of the normal oral cavity metabolome after the resumption of recommended oral hygiene practices. which suggests the potential for lingering harmful sequelae of gingivitis. This work represents a characterization of the human oral metabolome over 35 days, providing a high-resolution metabolomic landscape for understanding oral health during gingivitis and opening new opportunities for combating progressive periodontal disease.

Dr. Soundara Viveka Thangaraj (Viveka) is a postdoctoral fellow in the lab of Dr. Vasantha Padmanabhan in the Department of Pediatric-Endocrinology at the University of Michigan.

She received her PhD in Genomics and Oncology, from the University of Madras, India. Her research centers around the developmental programming effects of pre-natal exposure to environmental chemicals, on long-term metabolic health of the offspring, using omic tools.

In the current research, she has utilized a translationally relevant, real-life, low-dose, chronic chemical exposure model, in which sheep are exposed to biosolids which is a source of real-life mixture of environmental chemicals.

Osagie K. Obasogie is the Haas Distinguished Chair and Professor of Law at the University of California, Berkeley School of Law with a joint appointment in the Joint Medical Program and School of Public Health. He received his B.A. in Sociology and Political Science (with distinction in both majors) from Yale University, his J.D. from Columbia Law School where he was a Harlan Fiske Stone Scholar, and his Ph.D. in Sociology from the University of California, Berkeley, where he was a fellow with the National Science Foundation. Abstract:
Exercise capacity, a central factor in determining quality of life in healthy aging as well as cardiovascular disease, is determined by a systems-level interaction of factors that are intrinsic to the heart and myocardium and factors that are extrinsic to the heart. Cardiac intrinsic factors include the metabolic power supply and mechanical pumping power of the myocardium. Extrinsic factors include the capacity of the peripheral vasculature to vasodilate in response to increasing demands of exercising musculature and the autonomic chemo- and baroreflexes. 

     We have developed a whole-heart and whole-body cardiovascular modelling framework to capture and test hypotheses on how specific myocardial, autonomic, and vascular mechanisms determine physiological limitations to cardiac power and output reserve and contribute to diminished left-ventricular power output (LVPO) and exercise intolerance in heart failure. Simulations are driven by a multi-scale model of myocardial metabolism and mechanics. Simulations based on a model parameterization representing a healthy young subject may be used to explore, from a theoretical perspective, if and how myocardial ATP supply can affect myocardial mechanics and power reserve. As an emergent property, the integrated model accurately predicts the oxygen cost of contraction during rest and exercise, yielding the correct quantitative relationship between mechanical power output (in the in vivo whole-body setting) and myocardial mitochondrial oxidative phosphorylation (reflected in myocardial oxygen demand). Simulations also reveal that, under physiological conditions, the myocardial capacity to synthesize ATP at a free energy level needed to drive myocardial mechanics reaches its maximal value at maximal exercise. Thus these theoretical predictions are consistent with the concept that the myocardial energy supply capacity limits physiological cardiac power capacity.

Bio:
Dr. Beard is a Professor in the Department of Molecular and Integrative Physiology and holds affiliate appointments in Biomedical Engineering and Emergency Medicine. His laboratory is focused on systems engineering approaches to understanding the biophysical and biochemical operation of physiological systems. Dr. Beard is the Director of the Virtual Physiological Rat (VPR) project, supported as an NIH National Center for Systems Biology, working to analyze, interpret, simulate, and ultimately predict physiological function in health and disease.

Dr. Beard earned an MS in Applied Mathematics and PhD in Bioengineering in 1997 under the supervision of Dr. James Bassingthwaighte at the University of Washington. After completing his PhD he conducted postdoctoral research at the Courant Institute of New York University, and as a Howard Hughes Medical Institute Postdoctoral Fellow. Before joining the University of Michigan, he was Professor of Physiology at the Medical College of Wisconsin.

The LSO will return to the stage of Hill Auditorium on Friday, Jan. 20, 2023 with Gustav Mahler’s Symphony No. 1, also called the “Titan” symphony, and two works by Wolfgang Amadeus Mozart: Ballet Music from Idomeneo, and his Symphony No. 1 in E flat major, K. 16.

The LSO will once again perform under the baton on Yeo Ryeong Ahn, who holds the Gilbert S. Omenn Music Directorship and is a second-year doctoral student in orchestral conducting at the U-M School of Music, Theatre & Dance. She will be joined by assistant conductor Luca Antonucci, a first-year doctoral student.

All LSO concerts are free and open to the public. The Jan. 20 performance will begin at 7:30 p.m. and will follow all COVID-19 protocols in effect at the time for performers and audience members.

In keeping with its role as an orchestra for members of U-M’s medical, health and life science community, the LSO may ask for more stringent protocols than other events.

Indigenous people still constitute <1% of participants in precision and genomic medicine research despite endeavors to increase inclusivity. Past ethical issues related to Indigenous genomics have not been adequately reconciled and are now being repeated in the new era of Big Data. Concerns persist about the collectivization of Indigenous data into open-access databases that circumvent tribal research oversight, the underestimation of socioeconomic and cultural factors contributing to health disparities, and continued biocommercial exploitation of Indigenous biomarkers.

Dr. Tsosie will describe community-engaged research and describe paths forward that center Indigenous people as the agents of access for their own genomic and health data. The future of Indigenous genomics is not mere inclusion but through recognition of Indigenous genomic and data sovereignty.

Abstract:

Most disease associated genomic variants have relatively modest effects on target gene expression in reporter or CRISPR perturbation assays. In addition, enhancer disruption in vivo often has surprisingly weak phenotypic consequences. I will present machine learning (ML) methods (gkm-SVM and DNN) which we use to learn the complex transcription factor combinations that control enhancer activity and cell fate. I will then use these methods to develop a quantitative model for enhancer activity which shows that while promoter knockdown has robust effects on target gene expression, individual enhancer knockdown is often weaker and affects temporal transition dynamics, but not the final steady state. This model provides an explanation of the paradox of how enhancer variation can be strongly associated with disease risk while having individually weak effects, by showing in detail how gene regulatory networks control developmentally important and disease relevant cell state transitions and cancer.

Genome sequencing has revealed that filamentous fungi contain many secondary metabolite gene clusters. Our labs have been actively engaged in chemically characterizing the metabolites produced by Aspergillus species using genome mining approaches. This talk will present our recent data characterizing secondary metabolites in the human pathogen Aspergillus fumigatus. I will also introduce our new sustainability efforts in developing polyethylene and polystyrene substrates as sole carbon sources for filamentous fungi. We have been able to produce fungal natural products using plastics retrieved from the Great Pacific Garbage patch.

Clay C.C. Wang, Ph.D.

University of Southern California

Clay Wang is a professor at the USC Alfred E. Mann School of Pharmacy and chair of the Department of Pharmacology and Pharmaceutical Sciences. He also holds a joint appointment in the Department of Chemistry.

Wang received his BA in chemistry from Harvard University in 1996 and a PhD in chemistry from the California Institute of Technology in 2001. After completing a two-year postdoctoral fellowship in chemistry and chemical engineering at Stanford University, he joined the faculty in the Department of Pharmaceutical Sciences at USC as an assistant professor in 2003.

His research program focuses on the interface of chemistry and biological sciences. Specifically, his lab has been studying the use of Aspergillus nidulans as a general host for the production of fungal natural products. The lab is exploring using the fungal host as a platform for upcycling of plastics into high value natural products.

The Antoniewicz laboratory for Metabolic Engineering and Systems Biology develops next generation tools and techniques for analyzing, engineering and manipulating microbial and mammalian systems applied to specific problems in biotechnology and medicine.

Maciek R. Antoniewicz is a Full Professor of Chemical Engineering at the University of Michigan. Dr. Antoniewicz earned his B.S. and M.S. degrees in Chemical Engineering from Delft University of Technology (2000), and his Ph.D. in Chemical Engineering from the Massachusetts Institute of Technology (2006). After graduating he performed post-doctoral research at the DuPont Company. Dr. Antoniewicz started as an Assistant Professor in 2007 at the University of Delaware and was promoted to Associate Professor in 2013 and to Full Professor in 2017. In 2019, Dr. Antoniewicz moved to the University of Michigan.

Dr. Antoniewicz is an expert and a pioneer in the field of 13C-metabolic flux analysis (13C-MFA). Dr. Antoniewicz has received many awards for his research accomplishments, including the DuPont Young Professor Award (2008), the James E. Bailey Young Investigator Award in Metabolic Engineering (2008), the NSF CAREER Award (2011), and the Biotechnology and Bioengineering Daniel I.C. Wang Award (2015). In 2018, Dr. Antoniewicz was elected as a Fellow of the American Institute for Medical and Biological Engineering (AIMBE). His current interests include elucidating syntrophic interactions in microbial communities, adaptive laboratory evolution, optimizing CHO cell cultures for therapeutic protein production, and metabolic engineering of microbes for enhanced utilization of renewable substrates for production of value-added chemicals.

Abstract

One of the most important and difficult to quantify interactions in microbial communities is the cross-feeding of nutrients between community members. Experimental approaches that are based on metabolic measurements cannot precisely identify which metabolites are exchanged between community members. To address this challenge, we have developed a 13C-metabolic flux analysis technique that can precisely pinpoint exact metabolite that are being cross-fed. In this presentation, I will demonstrate applications of our new 13C-flux analysis approach to elucidate metabolite exchanges in several microbial co-cultures. To determine which metabolites are cross-fed (and at what rates), cells are cultured in parallel in the presence of carefully selected isotopic tracers. Isotopic labeling is then measured by mass spectrometry and the data is then analyzed using our new 13C-flux analysis approach. Using statistical methods, we can then precisely identify what metabolites are exchanged by the different strains, in which direction the cross-feeding occurs, and what the rates are for these cross-feeding fluxes. As an example, in E. coli co-cultures we identified nine metabolites that are consistently cross-fed. These include organic acids, intermediates of the glycolysis pathway, intermediates of the citric acid cycle, and some (but not all) amino acids. To verify these predictions, additional 13C-tracer experiments were conducted and complemented with 13C exo-metabolomic measurements. Taken together, these examples demonstrate that our new analysis techniques can significantly improve our understanding of metabolic interactions in microbial communities (which cannot be resolved with current approaches), and in the future will enable rational engineering of microbial interactions and assembly of microbiomes for practical applications in medicine and biotechnology.

Abstract:

Microbiomes have been proposed to regulate many aspects of animal physiology and dysregulated microbiomes are known to contribute to numerous pathologies in humans. Despite these findings, many of the molecular mechanisms by which bacteria can influence host physiology remain unknown. In an effort to identify novel molecular mechanisms by which bacteria can promote normal animal physiology my lab has established a high-throughput screening platform using the model animal C. elegans that can screen 1,500 individual bacterial isolates per week for effects on animal physiology. Using this platform, we have now identified multiple new species of bacteria that restore normal physiology in mutant models of human disease. Furthermore, using transposon screening, we have begun to uncover some of the mechanisms bacteria use to mediate these effects on animals. In the long-term we aim to test if such mechanisms are evolutionarily conserved in bacteria that colonize human microbiomes or if such mechanisms can be adapted to treat human disease.

BIOGRAPHY:

Dr. Nick Burton explores how chromatin remodeling contributes to human disease and how a person’s environment can impact their health and the health of their offspring. Recently, this work has become particularly interested in the diverse ways microbes and the microbiome can influence these processes. He earned a B.S. in biology from University of Wisconsin-Madison, where he worked in the labs of both Dr. Anna Huttenlocher and Dr. Scott Kennedy. He was then awarded a graduate research fellowship from the National Science Foundation (NSF) and earned a Ph.D. in biology from Massachusetts Institute of Technology, where he studied under the mentorship of Dr. H. Robert Horvitz. As part of his dissertation work, Dr. Burton sought to develop new paradigms to study the mechanisms by which parental environment regulates offspring physiology. In 2017, he joined the Centre for Trophoblast Research at University of Cambridge as an independent Next Generation Fellow. While there, he investigated how environmental bacteria can affect development, physiology, metabolism and neuronal function of individuals and their offspring. In 2021, he joined Van Andel Institute as an assistant professor in the Department of Epigenetics.

Abstract:

Microbiomes have been proposed to regulate many aspects of animal physiology and dysregulated microbiomes are known to contribute to numerous pathologies in humans. Despite these findings, many of the molecular mechanisms by which bacteria can influence host physiology remain unknown. In an effort to identify novel molecular mechanisms by which bacteria can promote normal animal physiology my lab has established a high-throughput screening platform using the model animal C. elegans that can screen 1,500 individual bacterial isolates per week for effects on animal physiology. Using this platform, we have now identified multiple new species of bacteria that restore normal physiology in mutant models of human disease. Furthermore, using transposon screening, we have begun to uncover some of the mechanisms bacteria use to mediate these effects on animals. In the long-term we aim to test if such mechanisms are evolutionarily conserved in bacteria that colonize human microbiomes or if such mechanisms can be adapted to treat human disease.

BIOGRAPHY:

Dr. Nick Burton explores how chromatin remodeling contributes to human disease and how a person’s environment can impact their health and the health of their offspring. Recently, this work has become particularly interested in the diverse ways microbes and the microbiome can influence these processes. He earned a B.S. in biology from University of Wisconsin-Madison, where he worked in the labs of both Dr. Anna Huttenlocher and Dr. Scott Kennedy. He was then awarded a graduate research fellowship from the National Science Foundation (NSF) and earned a Ph.D. in biology from Massachusetts Institute of Technology, where he studied under the mentorship of Dr. H. Robert Horvitz. As part of his dissertation work, Dr. Burton sought to develop new paradigms to study the mechanisms by which parental environment regulates offspring physiology. In 2017, he joined the Centre for Trophoblast Research at University of Cambridge as an independent Next Generation Fellow. While there, he investigated how environmental bacteria can affect development, physiology, metabolism and neuronal function of individuals and their offspring. In 2021, he joined Van Andel Institute as an assistant professor in the Department of Epigenetics.

Bio:
As the Vice President of R&D - Biologics, Dr. Lakmal Jayasinghe oversees all biological research and development projects at Oxford Nanopore. Lakmal joined ONT in 2006 after finishing his PhD in chemical biology in the University of Oxford. During his PhD in the Hagan Bayley group, Lakmal has studied different nanopores and has gained a wealth of knowledge in engineering nanopores using genetic and chemical approaches. His responsibilities at ONT include improving the readout signal of ONT platforms by upgrading its current nanopore reader and motor, as well as discovering new versions of nanopores, motors and chemistries to suit various ONT applications including protein sequencing. Lakmal also works with many academic collaborators across the world to ensure that Oxford Nanopore uses the best possible biological components and chemistries in its platforms.

Please join us in-person for a seminar highlighting environmental research. Marc Weisskopf's research interests mostly come from his background in neurobiology. He is interested in how environmental factors affect the nervous system, as well as the epidemiology of neurologic disorders. Current areas of his work include how environmental exposures relate to:
* Autism Spectrum Disorders (ASD)
* Attention Deficit/Hyperactivity Disorder (ADHD)
* Mental health
* Cognitive function/Alzheimer’s disease (AD)
* Amyotrophic lateral sclerosis (ALS)
Some examples of his current work include exploring how exposure to toxicants (e.g., lead, manganese, and air pollution) affect cognitive function and psychiatric symptoms; how air pollution and other toxicants relate to autism spectrum disorder (ASD); and how toxicant exposures like formaldehyde and lead relate to the development of amyotrophic lateral sclerosis (ALS). In addition, he has a large study in Israel and Denmark to explore the relationship of currently used medications to ALS. He is also very interested in issues of epidemiological methods and causal inferences as they relate to his work.

Abstract:
In 1996, Dr. Shilatifard identified the biochemical function of the Mixed Lineage Leukemia (MLL) gene translocation partner for the first time. The translocation partner, a gene called ELL, encodes an elongation factor: a protein that controls when, where and how fast the RNA Polymerase II transcription machinery can “read out” the instructions encoded in our genes. This discovery began a scientific journey to better understand elongation factors and other transcriptional control mechanisms, both in embryonic development and in cancer. Taking you along on this journey, Dr. Shilatifard will describe how he isolated all of the other MLL translocation partners in a “tour de force” application of biochemistry and molecular genetics, resulting in the astounding finding that these seemingly unrelated proteins actually function together within the (now widely known) Super Elongation Complex (SEC). In further research, the SEC emerged as a central factor regulating gene transcription at the elongation stage, and the lab has developed chemical compounds to disrupt or inhibit the Super Elongation Complex that are now in pre-clinical studies for the treatment of cancers caused by transcriptional defects. The story continues with his lab’s current research investigating how elongation factors, chromatin and the epigenome control transcription in human development and disease.

Bio:
Dr. Ali Shilatifard, Chairman and Professor of Biochemistry and Molecular Genetics and Professor of Pediatrics at Northwestern University Feinberg School of Medicine in Chicago, graduated from Kennesaw State University in 1989 with a degree in Chemistry with an emphasis in Organic Chemistry. While at KSU, Shilatifard slept in his car at night for a period in front of the gym and worked all over the campus from bussing tables at the student center, to cooking in the back kitchen, serving as the dispatch for the Kennesaw State College police department, and working as a TA in the Chemistry lab and Math lab to support himself and his education. He was supported by a full academic scholarship by KSU after his first year and graduated in four years. Now, Shilatifard is a renowned biochemist and cancer biologist. He made a seminal contribution to the field of leukemia biology early in his career by identifying the function of a gene translocation in childhood leukemia for the first time. In the 25 years since that discovery, he has dedicated his career to revealing the causes of childhood leukemia and other cancers and to leveraging these findings for the development of new cancer therapies. He currently runs an active research laboratory, heads a large department while he also is the Director of the Simpson Querrey Institute for Epigenetics. Shilatifard has authored over 250 scientific publications and is one of the founding members and is the current Editor of the online publication, Science Advances. He is one of the few scientists in the United States funded by the National Cancer Institute’s Outstanding Investigator Award, and in an effort to inspire young students to consider the STEM fields, he and his wife Laura started the Simpson Querrey Inspire Program.

The disproportionate impacts of disasters for communities of color have been documented repeatedly for both natural and human-caused disasters. In Michigan:
* More than 500,000 Michigan residents live within one mile of a facility storing large amounts of extremely hazardous chemicals;
* Those residents are disproportionately Black, Indigenous and People of Color (BIPOC) and confront daily risks of spills, explosions, and other environmental disasters at the facilities, as well as from the transport of chemicals to and from them through their communities;
* Children of color are more than two times as likely as white children in Michigan to live in the shadow of facilities housing hazardous substances.

This event, organized by Detroit community leaders with support from local academic institutions, will:
* Discuss why emergency preparedness and response is an environmental and climate justice issue;
* Describe gaps and challenges in Detroit’s emergency preparedness and response, from and environmental and climate justice perspective;
* Share recommendations for improving emergency preparedness and response for environmental and climate justice communities.

Anna Young, MS, PhD is a research associate in the Department of Environmental Health and Associate Director of the Healthy Buildings Program at the Harvard T.H. Chan School of Public Health. Her research focuses on our indoor exposures to complex mixtures of hormone-disrupting chemicals and builds evidence for healthier materials as a strategy to reduce these chemical exposures in buildings. She earned her PhD and MS in Environmental Health from the Harvard Chan School, and she also holds a BA in Computer Science and Environmental Studies from Yale University.

ABSTRACT:

Genomic rearrangements, also known as structural variations (SVs), are large scale alterations that changes the DNA structure. They include deletions, duplications, insertions, and other forms that are accompanied by copy number changes as well as inversions, translocations, and other copy-neutral forms. They are an important type of variation, affecting an order of magnitude more base pairs than single nucleotide variations (SNVs) in normal human population. In cancer, several chromosomal translocations have been identified and subsequently became targets of successful treatments. However, the functional impact of genomic rearrangements and their roles in treatment response are largely unexplored. We are developing new computational methods and exploring large scale cancer omics data to infer the mutational mechanisms leading to these alterations, to identify potential disease-driving events, and to study how they affect treatments.

Abstract:

From the initial genetic patterning to the assembly of the whole embryo, how ordered structures form has always been a fundamental question in developmental biology. The recent advances in imaging technologies are now providing us with direct visual access to the development of whole organisms in unprecedented detail. However, it is clear that to make the best use of this complex data, we need the help of computer algorithms that can mine it for biologically relevant information, summarize and then visualize it for us. The first project I will present focuses on the mechanisms driving the development of diverse 3D bone morphologies in mice. There, we developed algorithms to analyze micro-CT images to deconstruct the final morphology of each bone into the contributions made by remodeling of mineralized tissue vs. the cartilaginous growth plates. This allowed us to demonstrate the central role of the growth plate in 3D bone morphogenesis in normal and pathological development. The second project aims to understand how different cell activities contribute to large-scale morphogenetic movements. To this end, we generated the first whole embryo single-cell atlas of morphogenetic activity underlying gastrulation in the fruit fly, then used these maps to derive a statistical model associating cell shape changes and rearrangements to body axis elongation.

Areas of Interest:

My lab focuses on understanding how cells organize to form various tissue morphologies during normal and pathological development. We use two model systems to achieve this: the musculoskeletal system in mice and whole embryo development in fruit flies. Our approach centers on the development of cutting-edge bioimage informatics and data science algorithms to analyze the dynamics and the underlying patterns in developing tissues through 3D and 4D (3D+time) fluorescence images.

Neural progenitor cells in both mammals and insects transit through multiple states of competence to produce diverse neural cell types, losing the ability to generate early-born neurons while gaining competence to generate later-born ones. How is competence regulated to ensure the development of a highly organized structure like the brain? Using Drosophila, we find that neuroblasts (fly neural progenitors) undergo a developmentally-timed reorganization of their genome that impacts competence to activate genes in the descendent neurons. Here I will discuss mechanisms of neuroblast genome reorganization and the role of nuclear proteins in competence regulation underlying neural diversification.

Drawing from their decades of experience, from conservation to COVID reporting, Liz Neeley and Ed Yong will discuss the art and science of sense-making in a time of crisis, and why it's crucial to the challenges that we face.

Over the past fifteen years, she has specialized in environmental science, journalism, policy and social media, always searching for the best ways of spreading ideas through complex networks.

**Ed Yong** is a science staff writer with The Atlantic. He was awarded the 2021 Pulitzer Prize for Explanatory Reporting for his crucial coverage of the coronavirus pandemic, and was named “the most important and impactful journalist" of 2020 by Poynter. He anticipated the course of the virus, the complex challenges that the U.S. faced, and the government’s disastrous failure in its response. He is the author of two New York Times bestsellers, An Immense World and I Contain Multitudes. Before he joined The Atlantic, Ed’s writing was featured in National Geographic, The New Yorker, Wired, The New York Times, Nature, New Scientist, Scientific American and other publications.

A homeostatic control system for breathing regulation serves to maintain blood gases and tissue acid-base status, with lung ventilation dynamically regulated to maintain CO2/H+ within a narrow physiological range. In this talk, we will describe the identification of a long-sought group of brainstem neurons that are critically responsible for CO2 sensation in the service of breathing control, and of the molecular proton detectors that subserve this sensory function. We will also describe some of the intrinsic properties and neuromodulatory capabilities of these neurons, including our discovery of a peptidergic system that activates at the time of birth to stabilize breathing. Finally, we will provide evidence that this system may contribute to various breathing disorders, including congenital central hypoventilation syndrome (CCHS).

Doug Bayliss, Ph.D.
Joseph & Frances Larner Professor and Chair of Pharmacology

Doug Bayliss obtained his Ph.D. in Physiology from the University of North Carolina and, after postdoctoral training in Physiology and Biophysics at the University of Washington, he joined the faculty at the University of Virginia where he is currently the Joseph and Frances Larner Professor and Chair of Pharmacology. Dr. Bayliss has a long-standing interest in cellular and ionic mechanisms that regulate brain function, with a particular interest in the receptors, signaling pathways and ion channel effectors that underlie neurotransmitter/neuropeptide modulation of brainstem neurons involved in cardiorespiratory control. Another area of interest is in understanding activation of Pannexin ion channels, which are widely expressed and release ATP and other metabolites for intercellular signaling in multiple (patho)physiological contexts.
