SUMMARY:Livestream / Virtual:New Urban Mobility: Transformations and & Value Creation
DESCRIPTION:Currently\, urban mobility is facing several challenges: some are the result of long-term urbanization trends and others are the outcome of more recent changes to population behaviors. Addressing these challenges requires systems thinking and a broadening of collaboration between automakers\, mobility services companies\, and cities. These constituencies must transform\, which will determine their new roles in the new\, urban mobility. New value chains will emerge along with novel forms of value that will be realized through innovative business models. This presentation\, brought to us by Evangelos Simoudis\, Ph.D.\, will highlight the decisions automakers\, mobility service companies\, and cities must make\, and the transformations they will undergo with regards to technology and business models. Dr. Simoudis will also discuss the risks arising from the use of data and artificial intelligence in new\, urban mobility.

SUMMARY:Livestream / Virtual:The Clements Bookworm: Detroit’s Hidden Channels\, French-Indigenous Families in the 18th Century
DESCRIPTION:Dr. Karen Marrero discusses her new book *Detroit’s Hidden Channels: The Power of French-Indigenous Families in the Eighteenth Century* (2020). \n\nFrench-Indigenous families of the 18th century were a central force in shaping the history of Detroit. Situated where Anishinaabe\, Wendat\, Myaamia\, and later French communities were established and where the system of waterways linking the Atlantic Ocean to the Gulf of Mexico narrowed\, Detroit’s location was its primary attribute. While the French state viewed Detroit as a decaying site of illegal activities\, the influence of the French-Indigenous networks grew as members diverted imperial resources to bolster an alternative configuration of power relations. Women furthered commerce by navigating a multitude of gender norms of their nations\, allowing them to defy the state that sought to control them by holding them to European ideals of womanhood. By the mid-18th century\, French-Indigenous families had become so powerful\, incoming British traders and imperial officials courted their favor. These families would maintain that power as the British imperial presence splintered on the eve of the American Revolution.\n\nKaren Marrero is a past Clements Library research fellow and a professor at Wayne State University.\n\nRegister at myumi.ch/gjgzR\n\n*This episode of the Bookworm is generously sponsored by Douglas Johnson.*\n\n*The Clements Bookworm is a webinar series in which panelists and featured guests discuss history topics. Recommended books\, articles\, and other resources are provided in each session. Inspired by the traditional Clements Library researcher tea time\, we invite you to pull up a chair at our [virtual] table. Live attendees are encouraged to post comments and questions\, respond to polls\, and add to our conversation and camaraderie.*

SUMMARY:Exhibition:Mary Sibande
DESCRIPTION:Sophie/Elsie is part of a series in which South African artist Mary Sibande (b. 1982) explores her family’s history. Three generations of Sibande women were employed as domestic workers in Apartheid South Africa\, which Sibande references through Sophie’s blue and white uniform—a dress synonymous with the profession there. Though not quite part of the born-free generation (the name given to children born after the start of Nelson Mandela’s presidency in 1994)\, Sibande was able to dream of life choices that were inaccessible to her ancestors. These dreams are acted out by her alter-ego Sophie\, a life-size fiberglass figure cast in the artist’s own likeness. Sibande created Sophie/Elsie in 2009 in honor of her great-grandmother\, who was given the Western name Elsie because her masters couldn’t be bothered to learn her African name. While closing her eyes\, Sophie imagines herself as an orchestra conductor\, as a superhero\, as a businesswoman. Here\, her maid’s uniform transforms into the dress of a Victorian queen\, complete with billowing cape and dramatic train. \n \nAt a time when indoor visits are limited\, UMMA is proud to be able to share Sophie/Elsie with our community. Eventually\, she will take center stage in a reinstallation of the permanent African galleries\, opening in fall 2021. The reinstallation will feature UMMA’s historical collection of African art\, as well as recent acquisitions of contemporary artworks from artists based in Africa and in the diaspora. \n \nThis museum purchase was made possible by Joseph and Annette Allen. \n\n

SUMMARY:Exhibition:A Garden of Earthy Delights: Ceramic Sculptures by Ann Arbor Potters Guild at Matthaei Botanical Gardens
DESCRIPTION:Inspired by the synergy between nature’s beauty and complexity\, and in partnership with the University of Michigan Matthaei Botanical Gardens\, artists from the Ann Arbor Potters Guild created work centered around Michigan’s natural landscapes and specifically for the Great Lakes Garden at Matthaei. Potters Guild artists created eight free-standing sculptures using the full range of traditional ceramic methods and tools\, the potter’s wheel\, press molds\, coil building\, slab construction\, and sculpting. Depictions range from naturalistic and representational\, to abstract\, as well as a whimsical nod or two to the timeless role of fantasy and myth in humankind’s communion with nature. Exhibit is free but scheduled reservations are required to visit the botanical gardens. Use this link to book your visit: https://www.signupgenius.com/go/8050f4aadad22a2fd0-matthaei1

SUMMARY:Performance:CANCELLED: Junior Brown - Rescheduled from 7/12/2020
DESCRIPTION:If you haven't seen Junior Brown yet\, you need to \"guit with it\"! Junior Brown is a one-man musical hot fusion machine\, and he's coming to town with his guit-steel\, a unique hybrid contraption that's part electric guitar and part country steel guitar. People call him a cross between Jimi Hendrix and Ernest Tubb. But there's a lot more to say about Junior Brown. There's the fact that he's just about the best surf guitarist around\, for example. Or that he's a wonderful composer of traditional country songs\, the kind in which each outrageous pun leads you deeper into an emotional blind alley. Come check out the performer voted #1 House Rocker by the readers of Guitar Player! Come pick up a copy of Junior's recent album \"Deep in the Heart of Me.\"

Depictions range from naturalistic and representational\, to abstract\, as well as a whimsical nod or two to the timeless role of fantasy and myth in humankind’s communion with nature. SUMMARY:Performance:Postponed - Electric Hot Tuna: 50 Years - Rescheduled from 7/22 and 7/23/20
DESCRIPTION:Hot Tuna\, the duo of Jorma Kaukonen and Jack Casady\, perform with a well-honed and solid power—always in the groove from their years of experience and mutual inspiration. Started as a side project during Jefferson Airplane days\, the constant\, the very definition of Hot Tuna\, has always been Jorma Kaukonen and Jack Casady. The two boyhood pals have never wavered in one of the most enduring friendships in rock history. From their days playing together as teenagers in the Washington\, DC\, area\, through years in San Francisco (they're 1996 Rock and Roll Hall of Fame inductees)\, to their current acoustic and electric blues sound\, no one has more consistently led American music for the last 50 years. At the 2016 Grammys\, Jorma Kaukonen and Jack Casady were honored with Lifetime Achievement Awards\, and they're still coming up with new ideas. Hot Tuna performs an electric show tonight in observance of the band's 50th anniversary\, with drummer Justin Guip.

Despite successful first line therapies\, most patients relapse and develop more chemoresistant disease. This recurrence and development of chemoresistance is attributed to a rare population of tumor cells\, termed cancer stem cells (CSCs)\, which are more chemoresistant\, have the capacity to self-renew\, and can repopulate the entire tumor. Research has shown that CSCs are maintained by the non-cancer cells in the tumor microenvironment (TME)\, such as mesenchymal stem cells (MSCs)\, endothelial cells (ECs)\, and immune cells. Furthermore\, the role of non-cancer cells in clinical outcomes and chemoresistance has been highlighted by recent evidence showing that classification of HGSOC molecular subtypes\, which have variable clinical prognoses\, are influenced by the presence of non-cancer cells in the tumor. However\, it is currently unclear exactly how CSCs and the nuanced cell composition of the TME work together to promote chemoresistance. Current models used to study these phenomena either suffer from a lack of cellular complexity in the case of many in vitro models or impractical experimental constraints such as long latency periods and poor control over cell composition in patient-derived xenografts. To better understand the role of CSCs and the TME cells in chemoresistance\, practical in vitro model systems that more closely represent in vivo processes and microenvironments are needed. We hypothesize that the development of these in vitro model systems will contribute novel insights into TME-mediated CSC regulation and the development of chemoresistance in HGSOC.\n\nIn aim 1 we examined the emergence of chemoresistance in the context of CSCs by developing a 3D in vitro serial passaging model system that allows for long term culture of patient-derived tumor cells with periodic evaluation of stemness and chemoresistance. Using this model system\, we demonstrated increased proliferation\, expression of CSC markers\, tumorigenicity\, and chemoresistance over the course of long-term passaging\, reflective of emerging chemoresistance in vivo. Furthermore\, this system enabled us to define a malignant gene expression signature that is associated with chemoresistance\, tumorigenicity\, and stemness and to evaluate patient-specific chemoresistance development following treatment. Finally\, we demonstrated the translational value of this model system by showing that Metformin treatment can hinder CSC driven development of chemoresistance in a phase II clinical trial.\n\nIn aim 2 we developed a heterogeneous tumoroid culture system that enabled culture of patient-derived tumor cells with controlled ratios of MSCs\, ECs\, and immune cells to study TME-mediated maintenance of CSCs and chemoresistance. Using this model\, we found that changes in CD133+/-ALDH+/- CSC phenotypes in response to TME cells varied depending on the patient sample. We also observed increased tumorigenicity and chemoresistance in tumoroids compared to spheroids cultured with patient-derived tumor cells alone. Furthermore\, we found evidence of epithelial-to-mesenchymal transition (EMT) in tumoroids accompanied by altered CSC phenotypes and a malignant matrisome signature. All of this together supports idea that the non-cancer cells in the TME contribute to the development of advanced\, chemoresistant disease and implicates EMT\, changes in CSC phenotypes\, and matrix remodeling as the primary culprits.\n\nFinally\, in aim 3\, we utilized this tumoroid system to generate tumoroids with 23 different cell compositions to evaluate the role of TME cell composition in response to therapy. Drug assays with novel and traditional chemotherapies revealed that tumoroids with different compositions respond differently to therapy and that the number of monocytes included in the culture was associated with the greatest resistance to therapy. Furthermore\, our random forest models trained on the drug responses of each cell composition were able to predict drug response with moderate success. With these models we identified that nuanced differences in cell composition can influence drug response and that the strongest predictor of response to therapy was the total quantity of non-cancer cells. Overall\, this model demonstrates the potential of using the TME composition to predict patient drug response and direct clinical management.\n\nIn these aims we demonstrate the clear utility of complex and realistic\, yet practical in vitro model systems in the study of chemoresistance and CSC maintenance in ovarian cancer. Specifically\, we identified the link between CSCs and the development of chemoresistance in long term 3D in vitro serial passage culture. Furthermore\, we showed that the non-cancer cells in the TME can confer chemoresistance and promote EMT associated with altered CSC phenotypes and matrix remodeling. Lastly\, we demonstrated the potential of TME composition in predicting drug response. Overall\, the model systems presented in this study provide platforms that can be used to better understand the role of CSCs and the TME in chemoresistance and poor clinical outcomes. This could ultimately lead to the development of novel therapies\, enhanced clinical management\, and improved clinical outcomes.\n\nDate: Wednesday\, July 21\, 2021\nTime: 9:00 AM\nZoom: https://umich.zoom.us/j/96111622879\nPassword: 326862\nChair: Dr. Geeta Mehta
