DESCRIPTION:Inspired by the synergy between nature’s beauty and complexity\, and in partnership with the University of Michigan Matthaei Botanical Gardens\, artists from the Ann Arbor Potters Guild created work centered around Michigan’s natural landscapes and specifically for the Great Lakes Garden at Matthaei. Potters Guild artists created eight free-standing sculptures using the full range of traditional ceramic methods and tools\, the potter’s wheel\, press molds\, coil building\, slab construction\, and sculpting. Depictions range from naturalistic and representational\, to abstract\, as well as a whimsical nod or two to the timeless role of fantasy and myth in humankind’s communion with nature. DESCRIPTION:Collaborative science and the co-production of science involve working closely with partners at every stage - from conceptualizing a new project\, to conducting the research\, to refining tools to best meet a management need. The goal is to encourage mutually beneficial exchanges between researchers and resource managers. Essential to collaborative science is building relationships and engendering trust among the partners. NOAA’s NERRS Science Collaborative and RESTORE Science programs support collaborative science through funding and partnerships around protected and at-risk coastal and ocean areas. \n\nThis webinar\, the first jointly hosted event between the NERRS Science Collaborative and RESTORE programs\, will feature a panel discussion among three contributors to the programs\, highlighting important lessons learned and experiences on how to become effective co-producers of science.

DESCRIPTION:Diagnostic and monitoring devices based on protein biomarker measurements have the potential to inform courses of treatment in acute conditions and to help manage and prevent disease progression in chronic conditions. The importance of diagnostic and monitoring devices cannot be overstated as medicine is blind without them. However\, prevalent conditions such as traumatic brain injury (TBI) and chronic kidney disease (CKD) lack adequate diagnostic and monitoring tools. \n \nAssessment of TBI is typically done through neurological examination and neuroimaging techniques. While these methods can identify direct tissue damage to the brain\, they cannot assess the secondary damage stemming from the initial injury. The primary tissue damage sets off a cascade of secondary injuries\, such as neuronal cell death\, blood brain barrier breakdown\, edema\, and upregulation of inflammatory markers. Protein biomarkers have been proposed as a way of monitoring the progression of secondary TBI injury and of providing more sensitive diagnostic measures when used in conjunction with imaging and physical examination. However\, FDA authorized biomarkers\, glial fibrillary acidic protein (GFAP) and ubiquitin c-terminal hydrolase L1 (UCH-L1)\, can currently only be measured in a hospital laboratory. Analyzing TBI biomarker data in field and pre-hospital settings would enable rapid diagnosis and delivery of care\, so we developed a glass and silicon microfluidic device for quantification of TBI protein biomarkers via bead-based immunoassay. The device consists of a single channel with a variable height profile fabricated by slowly lowering a glass wafer into hydrofluoric acid. The device can capture and separate beads with diameters ranging from 1 to 5 micrometers. We also developed bead-based quantum dot-linked immunosorbent assays (QLISAs) for GFAP\, interleukin-6 (IL-6)\, and interleukin-8 (IL-8) by conjugating appropriate antibodies to 2.8\, 4.5\, and 1 micrometer diameter beads\, respectively. We used the variable height device to passively separate a mixture of assay beads with completed immunocomplexes into distinct bands where the diameter of the beads matched the height of the channel. The fluorescent intensity of each band was used to quantify a different biomarker concentration\, resulting in a customizable brain injury barcode. By changing out the bead size and antibodies used for each immunoassay\, the variable height device can keep pace with the developing field of TBI biomarker discovery and validation or be applied to biomarkers for another condition of interest. \n \nWe also developed an assay system to monitor CKD\, which is one of the ten most common chronic conditions in adults aged 65 and older. Clinical guidelines currently recommend that the urinary albumin-to-creatinine ratio (ACR) be measured once a year to monitor disease progression. However\, measuring the ACR at home can enable patients to self-identify the early signs of disease progression and consult their physicians\, potentially leading to improved outcomes. We designed and analytically validated inexpensive\, colorimetric assays for urinary albumin and creatinine intended to be used in a 3D-printed\, disposable microfluidic device. Discrepancies between the results from the colorimetric albumin assay and the hospital albumin assay when clinical urine samples were tested highlight the importance of incorporating analytical validation as a checkpoint in the development of microfluidic monitoring systems. \n \nOverall\, this work presents a novel method of multiplexing immunoassays in microfluidic systems that can be applied to measure protein biomarkers for TBI or another condition of interest and highlights the importance of integrating assay design with microfluidic device design to promote clinical translation.\n \nDate: Friday\, July 30\, 2021\nTime: 10:00 AM\nZoom: https://umich.zoom.us/meeting/register/tJYpdu2rrjsqH9LM0c4DZ8bs8Uj0BdggOEeE (pre-registration required)\nChair: Dr. Mark Burns

DESCRIPTION:Inspired by the synergy between nature’s beauty and complexity\, and in partnership with the University of Michigan Matthaei Botanical Gardens\, artists from the Ann Arbor Potters Guild created work centered around Michigan’s natural landscapes and specifically for the Great Lakes Garden at Matthaei. Potters Guild artists created eight free-standing sculptures using the full range of traditional ceramic methods and tools\, the potter’s wheel\, press molds\, coil building\, slab construction\, and sculpting. Depictions range from naturalistic and representational\, to abstract\, as well as a whimsical nod or two to the timeless role of fantasy and myth in humankind’s communion with nature. This potent combination\, paired with a robust tour calendar\, have made the veteran band hands-down favorites of fans and peers alike. From their base in Virginia’s Blue Ridge Mountains\, TSW have played the most prestigious festivals\, listening rooms\, and clubs in the world\, cementing a reputation as one of the top independent bands on the scene today.\n\nWith their 2020 release\, Everyone A Song\, Vol. 1\, The Steel Wheels demonstrate the humanizing power of honest storytelling\, setting the personal experiences of their supporters to song. \n\nWhat started as a ruined touring season turned into a time of great creativity\, as the band devised a new model of writing and recording. With the 5 members isolated from each other and their listeners\, they quickly shifted from veteran touring outfit to home recording gurus. Songwriter and lead singer Trent Wagler conceived of a project entitled Distance Together\, an avenue for fans of the band to commission works and send musical greetings to their loved ones. The initiative sparked a surprisingly impactful and intimate songwriting process\, with Wagler setting the personal stories of fans to song and verse. Songs were written for individuals\, friends\, and families\, many specific to a time and place. Some have been commissioned for weddings and anniversaries\, or to commemorate a lost loved one or the completion of a life's work. \n\nIn \"My Name is Sharon\"\, Wagler was tasked with memorializing a young woman and friend of the band\, lost at the age of 35. The resulting song remembers details of Sharon's life\, in all it's profundity and mundanity\, and demonstrates the power of remembrance held in saying someone's name. \n\nAlbum ender “Family is Power” follows the matriarch of a large family\, as she looks back on her long life and the family she loves\, now spread across the country. “She’s gonna see the truth in me\, in the golden hour. Come to the table one and all\, Family is Power.” In “The Healer\,” buoyant banjo and fiddle lines drive the story of a magnetic personality who\, after a lifetime of work and service is denied the opportunity to say goodbye to colleagues in the midst of the pandemic. “Water and Sky”\, a tribute to John Prine in its simplicity and depth\, paints pictures of a dreamy Kansas landscape\, and the strong personalities who hold witness to the ebb and flow of life. \n\nThe Steel Wheels have always been fiercely independent\, releasing albums under their own label\, with recording locations varying from a Virginia cabin\, to a rural Maine studio to various home studios. Now spread across 3 states with many miles between\, The Steel Wheels have invented an entirely new way of collaborating. Fortunately two of the members\, multi-instrumentalist Jay Lapp and percussionist Kevin Garcia were already skilled audio engineers with home studios at the ready. Others\, such as fiddle player Eric Brubaker\, who had to scrounge together a few mics from the touring van and an old laptop\, learning the ropes of digital audio recording on the fly. \n\nAccording to Brubaker\, \"Process has a great deal to do with the outcome of a creative undertaking. Some of the pressure is off when you are just at home\, instead of at an expensive studio\, so you might take some risks that you wouldn’t usually. Also\, we were working at a pretty fast pace\, so you just had to trust your first instinct and not overthink it.” \n\nDuring the first few months of the pandemic and cessation of all touring\, The Steel Wheels recorded over 60 songs in their isolated studios. Some covers\, some new editions of their own songs\, and some newly penned\, the creations generally started with Wagler recording a single instrument and vocals. The tracks were emailed to Garcia\, who layered in a backbone of percussion. Next in line was usually Brian Dickel who rounded out the rhythm section with upright and electric bass parts. \n\nPart by part\, the songs were arranged and recorded\, often with little discussion as to who was providing which part\, or even what instruments should be included. The band relied on their years of musical camaraderie and intuition to imagine the whole\, adding their voices while being careful to leave room for the next in line. The process yielded some exciting new sounds\, from the neo soul vocals and punchy bass lines of “Florida Girl” to the second line swagger of Garcia’s drums on “The Man Who Holds Up the World.\" \n\nThe Steel Wheels have been a band marked by their road tested friendship and desire to build community. 2020 would have marked their 8th year as hosts and curators of their Red Wing Roots Music Festival. Always a high-water mark of the year\, the 3 day festival brings together thousands of fans and friends every July to sing and sweat together in the Virginia sun. \n\nEveryone A Song\, Vol. 1 was born out of a desire to close the distance created in the midst of the Covid-19 shutdown. “Maybe I was dreaming of being an essential worker\,” says Wagler. “It made me ask the question: what’s essential about music? It’s the connection. The understanding. The beauty and magic of melodies that transcend us with the words that say\, ‘You there\, I see you\, and you see me\, and doesn’t it feel good to not be alone in the world?’ That’s what art has to offer. Especially right now. And planting real stories right in the middle of it kind of pushed aside any other pretense or distraction about being cool or whatever.” \n\nThis project has been further documented in an accompanying podcast\, We Made You a Song. Each episode dives into the creation of a single song on the album\, serving as an in-depth audio liner note. Wagler interviews the band as well as the actual subjects of each song to give a rare glimpse into the creative process\, and what it means for someone to have their experience distilled into melody and rhyme.

SUMMARY:Presentation:BME PhD Defense: Emine Sumeyra Turali-Emre
DESCRIPTION:Biomimetic nanoparticles (NPs) are bio-inspired inorganic nanoscale materials that replicate some biological nanostructures functionalities including self-assembly\, catalysis\, and enzyme inhibition. These functionalities are being investigated for and\, in some cases\, are being utilized in optics and electronics such as chemical sensors\, superhydrophobic coatings\, and antireflective surfaces. This thesis examines the utilization of biomimetic inorganic NPs for various problems in biomedical engineering.\n\nSpecifically\, in the first part of this thesis\, I address the problem on controversial explanations of the antibacterial and other biological activity of zinc oxide NPs that are frequently utilized in cosmetics\, textiles\, and biomedical fields. In the second part of the thesis\, I explore the self-organization of NPs into biomimetic supraparticles (SPs) for nucleic acid delivery that can be exploited as drug delivery agents.\n\nNPs have been used in the antimicrobial field for a long time\; however\, their antibacterial mechanism of action against different types of bacteria remains unclear and\, in many cases\, misinterpreted. Most of the studies on antimicrobial NPs suggest reactive oxygen species (ROS) formation\, ion release\, and membrane damage as the primary source of antibacterial activity. In Chapter 2\, we show that the mechanism of antibacterial activity for Staphylococcus aureus is remarkably more complex than generating ROS or the release of Zn2+ ions and is based on formation of biomimetic complexes of NPs with proteins.  Gene expression analysis demonstrated that ZnO-NPs significantly affect carbohydrate metabolism and cell energetics\, where the uridine monophosphate (UMP) biosynthesis pathway is highly upregulated. In Chapter 3\, we explore the ZnO-NP mode of entry into S. aureus and the cell metabolism. Here\, we showed that NPs enter the cells within 5 minutes of exposure and induce minimal membrane damage. We note that cells do not depolarize until 60 min post-NPs exposure. Thereby\, we highlight that membrane damage is not the primary mechanism of action but rather a downstream effect of ZnO-NPs exposure to bacterial cells. Taken together\, causing minimal ROS production and significant changes in carbohydrate metabolism and bioenergetics along with cell entry without immediate membrane damage imply the biomimetic function of these NPs. Further investigation into the antimicrobial mechanisms of biomimetic NPs is essential for future clinical translation. \n\nOver the past few decades\, there has been considerable interest in developing nanoscale constructs as effective delivery tools for high molecular weight drugs. In chapter 4\, I explore the self-assembly of NPs into compartmentalized SPs\, which mimics the structure of a virus to deliver nucleic acid into cells. The time-dependent self-assembly mechanism reveals that these SPs are formed from nanocup intermediates. We found that this intermediate stage is essential for the utilization of SP compartments. Nucleic acid is added to the system at this stage before SP formation\, and high encapsulation is achieved. Cantilina was introduced to bells during his university years\, when he played Cornell University's twenty-one bell chime. In graduate school\, Cantilina studied with Dave Johnson at the House of Hope carillon in St. Paul\, MN. In 2018-2019\, he studied carillon in Belgium with Koen Van Assche with financial support from the Belgian American Education Foundation. Cantilina lives in Cleveland\, OH\, where he performs frequently on the Alexander McGaffin Carillon.

DESCRIPTION:Inspired by the synergy between nature’s beauty and complexity\, and in partnership with the University of Michigan Matthaei Botanical Gardens\, artists from the Ann Arbor Potters Guild created work centered around Michigan’s natural landscapes and specifically for the Great Lakes Garden at Matthaei. Potters Guild artists created eight free-standing sculptures using the full range of traditional ceramic methods and tools\, the potter’s wheel\, press molds\, coil building\, slab construction\, and sculpting. Depictions range from naturalistic and representational\, to abstract\, as well as a whimsical nod or two to the timeless role of fantasy and myth in humankind’s communion with nature. DESCRIPTION:Type 1 diabetes is caused by the autoimmune destruction of insulin-producing β-cells\, resulting in a chronic metabolic disorder typically treated with exogenous insulin. Even with the aid of advanced insulin pumps and real time feedback systems\, blood glucose levels still deviate outside of the range maintained by native islets\, which places the individual at risk for vascular complications and life-threatening hypoglycemic events. Cell replacement therapies have demonstrated the capacity to tightly control blood glucose levels. The wide adoption of cell replacement therapy is hindered by limited availability of donor islets\, and the lack of effective methods to support the long-term function of these cells within a clinically accessible site. The results presented in this thesis address these limitations: through studying maturation of human pluripotent progenitor cell (hPPC) derived β cells within a transplantable biomaterial platform\, and evaluating novel approaches to the implantation and support of these cells during their continued maturation in vivo. \n\nFirst\, I present a study that examined delivery of hPPC-derived pancreatic progenitors within microporous PLG scaffolds into the epididymal fat pad\, the murine surrogate for the clinically relevant omental pouch. Kidney capsule injection\, the site most commonly utilized to test stem cell-derived β cell function in murine models\, was the comparison condition. We observed that the microporous scaﬀolds supported cell engraftment\, however the levels of circulating C-peptide were lower when compared to the kidney capsule condition. The scaﬀolds were subsequently modiﬁed to provide sustained release of exendin-4\, which led to signiﬁcantly increased C-peptide production. Image analysis revealed that exendin-4 releasing scaffolds enhanced the proportion of pancreatic progenitors that matured to monohormonal insulin producing cells.\n\nNext I present my findings from studying how hPPC-derived β cells mature and function within three transplantation sites: the i) scaffold delivery into the epididymal fat pad\, ii) scaffold delivery into the subcutaneous space\, and iii) the kidney capsule injection (control). Additionally\, we investigated the impact of blood glucose levels on maturation of the hPPC-derived β cells by transplanting mice with pre- or post-engraftment diabetes induction.  Hyperglycemia was ameliorated in the cohorts of mice that received scaffolds into the epididymal fat pad\, following a period of in vivo maturation.  The function of these cells was demonstrated by the reduction in blood glucose levels\, healthy increase in weight\, therapeutic levels of circulating human insulin\, and healthy responses to glucose challenge tests. The function from the epididymal fat pad was superior to the subcutaneous space and was observed to be comparable to the kidney capsule. No differences were observed in graft function between the cohorts whose grafts matured in a diabetic or non-diabetic environment\, yet several differences in gene expression were observed.\n\nMany of the current differentiation protocols culture the cells above a feeder layer in monolayer\, or in suspension within a bioreactor. Typically\, these protocols require the disruption of the cell niche during key differentiation stages or pre-transplantation handling. Biomaterial scaffolds maintain the integrity of cell-to-cell and cell-to-matrix connections by providing both a space for cell niche development as well as a vehicle for transplantation into the body. Herein\, I present results from testing the developmental stage in which progenitors are seeded into the 3D niche\, and two differentiation strategies prior to seeding: monolayer and suspension culture. Maturation was characterized via gene expression analysis\, glucose stimulated insulin secretion assay\, and nondestructive microscopy utilizing a sfGFP-C-peptide cell line that reports C-peptide production and secretion. We observed that seeding clusters during the key transition phase from pancreatic progenitor to pancreatic endocrine enhanced commitment to the final beta cell fate.\n\nThis work enhances our understanding of hPPC-derived beta cell manufacturing within scaffolds\, and delivery to an extrahepatic site to achieve normoglycemic blood glucose levels.\n\nDate:  Wednesday\, August 4\, 2021\nTime: 10:00 AM\nZoom: https://umich.zoom.us/j/98009467275\nPassword: betacell\nChair: Dr. Lonnie Shea

It was March of 2020\, and as the COVID-19 pandemic began tightening its deadly grip across the United States\, the roots rock stalwart found himself forced off the road for the first time in years\, stuck at home with a calendar full of cancelled tour dates and no one to perform for. Concerned by the chaos unfolding around him but certain he wasn’t the only one in search of some kind of solace\, Ezra decided to pick up his guitar\, turn on his phone\, and do what he’d always done in times of worry or doubt: play a show.  \n\n“I just started livestreaming from my house\,” Ezra recalls. “It was a way to connect with people\, to feel a little less lost\, a little less scared. I played and talked for about an hour\, and then at the end of the stream\, I told everybody\, ‘I’ll come back tomorrow if you come back tomorrow.’” \n\nEzra kept his promise and then some\, returning every single night after that for what he and his fans would eventually come to call The Gathering Series. Interactive and improvisatory\, the shows took shape on the fly\, drawing on daily life and frequently featuring newly written material and never performed songs from Ezra’s extensive back catalog. Through his own harrowing battle with COVID\, a move to the Mojave Desert\, a socially distanced summer tour\, and even his wedding day\, Ezra never missed a performance\, broadcasting without fail from wherever he found himself each night at 7pm Eastern. Driven by a belief that small grassroots communities have the power to impact the world\, Ezra - who has gone live daily since March 13th 2020 - has reached a cumulative audience of well over 15 million viewers worldwide\, a staggering feat for any artist that stands as a testament\, not only to his artistic endurance\, but also his ability to provide an accepting and supportive space for newcomers joining his tight-knit community of fans during a time when it’s needed more than ever. \n\n“The people who watch every night\, they’re tuning in for me\, but they’re also tuning in for each other\,” says Ezra. “They’re tuning in for comfort and compassion and connection\, but maybe also a sense of normalcy and grounding during this time that seems to be shaking the very foundations of who we’ve always thought we were. As special as The Gathering Series is\, I don’t think it could have achieved this kind of success if we didn’t have this incredible fan community around us that is inspired by engagement and activism.” \n\nWorking outside the confines of the traditional music industry\, Ezra and his band\, the Adam Ezra Group\, built that foundation through sheer grit and determination\, regularly performing hundreds of shows a year for the past two decades without any major label or mainstream radio support. Fusing folk intimacy and rock energy with soul power and pop charm\, the band first emerged from Boston in the early 2000s and quickly garnered widespread acclaim for their bold\, insightful songwriting and interactive\, euphoric performances. From hole in the wall bars and house concerts to sold-out headline shows and dates with the likes of Little Big Town\, The Wallflowers\, Gavin DeGraw\, Train\, and The Wailers\, Ezra and his bandmates treated every single gig like their last\, attracting a die-hard following that believed not just in the music\, but in their commitment to activism and social change\, as well. Charity events and fundraisers were a regular part of the group’s touring schedule\, and each year\, thousands of Adam Ezra Group fans would gather in Boston for the band’s homegrown festival\, The Ramble\, which raised money and marshaled resources to help homeless veterans find safe and dignified housing. \n\n“With the pandemic raging\, 2020 was going to be the first time in ten years that we couldn’t host a Ramble\,” says Ezra\, “so we decided to incorporate it into The Gathering Series and put on a 24-hour\, non-stop livestream. It was a crazy idea\, but by the end of it\, we had tens of thousands of people rambling with us from around the world and we’d broken our previous fundraising record\, collecting enough money to help 64 of our veteran heroes off the streets.” \n\nWhile that 24-hour livestream was undoubtedly the most taxing installment of The Gathering Series\, finding ways to broadcast a new\, unscripted hour of live music every night for a year was rarely easy. In April\, Ezra came down with COVID and found himself so weak he could barely get out of bed to perform. In May\, he and his wife traded Massachusetts for the Mojave\, setting up a temporary home base in a cabin outside Joshua Tree\, where Ezra would continue to stream daily from the desert. Over the summer\, he and the band cooked up a way to bring The Gathering Series to the real world\, performing nearly 50 safe\, socially distanced shows in parks and backyards around the country while still managing to go live every night at 7pm. \n\nApart from the emotional and physical commitment\, playing every night hasn’t been without its logistical challenges as well. “I remember one night I was in a hotel\, and the WiFi went out\,” says Ezra. “I jumped in the van and just started driving until I found service\, and then I pulled over and played the show right there on the side of the road from the driver’s seat. Turned out to be one of my favorite memories looking back on the year.”  \n\nAs if over 425 shows since the pandemic started isn’t enough to keep them busy\, Ezra and his bandmates—fiddler Corinna Smith\, drummer Alex Martin\, and bassist Poche Ponce—also embarked on an ambitious new recording series in 2020. Dubbed The Album Project\, the endeavor will see the band releasing at least 19 new singles over the next few years\, trading in the traditional LP format for a steady flow of fresh material designed to keep their ever-expanding community active and engaged even beyond the pandemic. The series opened up\, fittingly enough\, with “All Right Today\,” an effervescent\, feel-good ode to putting one foot in front of the other\, to living in the moment and leaning on one another to get through our hardest times. \n\n“I wrote that song years before COVID\,” says Ezra\, “but that line\, ‘We will be all right today\,’ kept coming back to me through this whole experience. It felt like a mantra I continue to lean on right now and the right way to kick this whole thing off.” \n\nIn celebration of their 365th Gathering show\, Ezra and the band followed that release up with The Album Project’s second installment\, “Switching To Whiskey\,” which hints at old-school country and Appalachian folk as it meditates on the power of heartbreak and memory. Like much of Ezra’s catalog\, the bittersweet track is intimate and organic\, fueled by stripped-down\, acoustic instrumentation and the kind of raw\, honest vocals that cut straight to the heart of things.  “Switching to Whiskey” has well over 225K streams on Spotify\, and a new music video on YouTube. \n\n“As soon as we started playing that one live\, it became one of our most-requested\,” says Ezra. “And the beauty of The Album Project is that when our fans are really connecting to something\, we can just record it and get it right out to them\, because they’re truly the engine behind everything we do.” \n\nThe third release from the project “Devil’s Kiss” released in May 2021\, and focuses on Ezra’s reaction to the Covid-19 pandemic\, and all of the challenges everyone has faced over the past year. \n\nWhat comes next for the Adam Ezra Group? They have started another socially distanced tour and have geared up to start a full routed tour again in June 2021. Perhaps another 24-hour livestream will be in the works? Perhaps something even crazier. The future is anyone’s guess right now\, and the only thing the Adam Ezra Group knows for sure\, is they’ll be out there making music however and whenever they can.
