While the goal of these studies is to design new and effective ways to treat disease and provide better understanding of ourselves as well as the world around us\, many are also taking the time to share the beauty of their work with others through a program called BioArtography. In the course of research\, scientists use special stains to add color to the otherwise transparent tissues. Microscopes then allow detailed observation of the tiny\, colorful biological structures revealed in these images. This results in a fascinating combination of art and science that U-M researchers are capturing in pictures taken through microscopes and turning into artworks that would look beautiful on any wall.\n\nThe goal of these studies is to design new and effective ways to treat disease and provide better understanding of ourselves as well as the world that surrounds us. Mother Cyborg draws from over 15 years experience as a community organizer in Detroit\, MI where individuals gained access and agency to (re-)build their neighborhoods\, and run their own Internet service providers. In this exhibition\, Nucera expands her artist self through fiber works where she addresses critical issues of surveillance\, data collection\, the redaction of love to likes\, and the complexity of identity within it all. \nMother Cyborg states\, “This work aims to return a sense of wonder and magic to all of us\, and rekindle the awe we may have first felt when using a geolocating star map\, or being able to video call a person across borders. It features iconic works from Weisberg’s artistic career - Waterborne (1973)\, A Year Passes (1985) and Questioning Veronese (2011) - where she explored the interconnectedness and continuity of successive generations. Anchored in themes of personal and familial memory\; ancestral knowledge embedded in her Jewish heritage\, and admiration for Western art history\, her unframed canvases\, and works on paper create liminal spaces that collapse the time and space\, where family\, friends from the present and past share space with great painters and Holocaust survivors and those who perished. The Shtetl\, A Journey and Memorial\, a limited- edition artist book that she wrote and illustrated comprising 9 intaglio prints in 1971\, lies at the heart of the exhibition. In it she paid homage to the small Jewish towns in Poland (where her ancestors are from) before the Holocaust. This significant work was a touchstone for her artistic and professional career as it propelled her move from Michigan to Los Angeles in 1969.\n \nBorn and raised in Chicago\, Weisberg moved to Ann Arbor to pursue her higher studies\, she completed her B.Ed (1964)\, after spending three years in Perugia\, Italy where she earned a Laurea di Belle Arti in 1962\, and MFA (1965). Since her arrival in Los Angeles\, Weisberg has been a formidable influence and mentor to decades of artists as an artist\, Professor of Fine Arts and former Dean at the University of Southern California Roski School of Art and Design. Weisberg was the Director of the USC Initiative for Israeli Arts and Humanities\, and the founder of the Jewish Artists Initiative of Southern California. Her first major survey in Los Angeles was in 1979 at the Los Angeles Municipal Art Gallery. She and Judy Chicago were the first two artists exhibited at The Women’s Building\, Los Angeles. Their solo exhibitions inaugurated that venue. Weisberg has had more than 80 solo exhibitions and nearly 200 group exhibitions internationally\, including a major exhibition at the Norton Simon Museum\, Pasadena and a retrospective\, at the Skirball Museum\, Los Angeles as well as a solo exhibition at the Huntington in San Marino. She is the recipient of many prestigious awards\, some notable ones are the Printmaker Emeritus Award from the Southern Graphic Council International in 2015 and the Foundation for Jewish Culture’s 50th Anniversary Cultural Achievement Award in 2011. She has been the recipient of the Art Leadership Award\, National Council of Art Administrators and the Women’s Caucus for Art Lifetime Achievement Award\, 2009. Beyond the usual descriptors such as the mean and variance\, quantile and superquantile (also known as the expected shortfall or conditional value-at-risk) regression can capture the differential covariate effects on the upper or lower tails of the response distribution. This dissertation studies some fundamental aspects of the statistical inference of quantile and super quantile regression. \n\nIn the first part of the dissertation\, we propose a novel approach to superquantile regression with a critical modification of an optimization formulation in the recent literature. Most existing approaches for superquantile regression rely explicitly on the modeling of the conditional quantile function. In this dissertation\, we offer new insights into an optimization formulation for the superquantile\, based on which we provide and validate a direct approach to superquantile regression estimation without relying on additional quantile regression modeling. Operationally\, the approach can be well approximated by fitting a linear quantile regression to an array of pre-estimated conditional superquantile processes. This approach achieves implicit weighting of the data\, which is found to be automatically adaptive to data heterogeneity in a variety of scenarios. With certain initial estimators based on binning of the covariate space\, we show the proposed superquantile regression estimator is consistent and asymptotically normal. Via theoretical and numerical comparisons\, we show that the proposed approach has competitive\, and often superior\, performance relative to other common approaches in the literature. \n\nIn the second part of the dissertation\, we study pseudo-Bayesian inference for possibly sparse quantile regression models. We find that by coupling the asymmetric Laplace working likelihood with appropriate shrinkage priors\, we can deliver pseudo-Bayesian inference that adapts automatically to the possible sparsity in quantile regression analysis. After a suitable adjustment on the posterior variance\, the proposed method provides asymptotically valid inference under heterogeneity. Furthermore\, the proposed approach leads to oracle asymptotic efficiency for the active (nonzero) quantile regression coefficients and super-efficiency for the non-active ones. We also discuss the theoretical extension when the covariate dimension increases with the sample size at a controlled rate. By avoiding the need to pursue dichotomous variable selection as well as nuisance parameter estimation\, the Bayesian computational framework demonstrates desirable inferential stability. The course is both hands-on and conceptual. It begins by discussing what is unique about web surveys and when their use is most appropriate\, followed by an introduction to survey errors that can affect the quality of web survey data. Small groups of students will each develop a research problem and a questionnaire to address their problem\, designed for online administration. They will pretest the question wording\, program the questionnaire using a web survey development platform (no programming experience is required)\, and assess users’ (respondents’) experience while interacting with the web-based instrument. Students will also develop basic plans for data collection and analysis. In Earth’s nightside magnetosphere\, the dynamics of electrons within the near-Earth magnetotail are highly dependent on variations in the solar wind and overall solar activity. The electrons are the source population for the Earth’s outer radiation belts\, contribute to storm-time ring current pressure and energy\, and precipitate into the ionosphere forming part of the aurora. Additionally\, this population of electrons are most responsible for the spacecraft surface charging environment at geosynchronous (GEO) orbit. Some dependencies of superthermal electrons on solar wind variations have been identified through several previous investigations\; however\, there is much that is not known\, and it has been difficult to predict their behavior.\n\nUtilizing more than twelve years of data from THEMIS spacecraft\, we have investigated the long-term and short-term dependencies of electron flux in the near-Earth plasma sheet to solar wind variations. On time scales of a solar sunspot cycle\, we explore the response of energetic electron flux in the near-Earth plasma sheet to solar wind and geomagnetic activity. We show that with only slight solar wind driving (solar wind flow speed\, VSW = 400–500 km·s-1) and weak geomagnetic activity (Auroral Electrojet index\, AE = 100–300 nT)\, there is a substantial increase in median 12–52 keV electron flux. We also explored the role that solar wind driving has on short-term (1–2 hours) electron flux variations and found that VSW is the most significant contributor to severe spacecraft surface charging environments at GEO. Furthermore\, simply the presence of elevated geomagnetic activity (as indicated by the AE index) is a sufficient indicator of risk for an extreme charging environment. In examining 101 observations of the change of electron flux in the near-Earth plasma sheet during substorms\, we failed to find a dependence of flux changes on substorm strength\, nor spacecraft location\, nor location relative to the peak of auroral activity.\n\n\nFinally\, we develop an empirical\, machine-learned neural network model of electron flux in the near-Earth plasma sheet\, dependent on inputs of solar wind parameters and their time history. Our model overcomes limitations of previous models by including only inputs that are external to the magnetosphere and predicting differential flux at a wide range of energies. We calculate several model–observation metrics—our model predicts electron flux with a Pearson correlation coefficient between 0.55–0.77 and has a median symmetric accuracy of between 41–140% (metric ranges depend on energy)\; and\, we rank which solar wind parameters are most relevant to predictions. We show that including short time (5-minute) resolution inputs to the model does not result in predicting small scale (1-hour) variations of plasma sheet electron flux. In this paper\, we propose a coded computing approach for recovering matrix inverse approximations. We first present an approximate matrix inversion algorithm that does not require a matrix factorization but uses a black-box least squares optimization solver as a subroutine to give an estimate of the inverse of real full-rank matrices. We then present a distributed framework for which our algorithm can be implemented and show how we can leverage from sparsest-balanced MDS generator matrices to devise inverse coded computing schemes. We focus on balanced Reed-Solomon codes\, which are optimal in terms of computational load\; and communication from the workers to the master server. We also discuss how our algorithms can be used to compute the pseudoinverse of a full-rank matrix and how the communication can be secured from eavesdroppers. The course focuses on case studies with specific large-scale national surveys: the National Comorbidity Survey-Replication (NCS-R)\, the National Health and Nutrition Examination Surveys (NHANES)\, and the Health and Retirement Study (HRS). Relevant design features of the NCS-R\, NHANES and HRS include survey weights that take into account differences in probability of selection into the sample and differences in response rates\, as well as stratification and clustering in the multistage sampling procedures used in identifying the sampled households and individuals. After introducing essential concepts related to complex sample designs\, the course will turn to the construction of survey weights\, estimation of sampling variance\, descriptive analysis\, regression analysis\, and finally special topics in the analysis of survey data. Despite their promise\, there are no approved agonist antibodies for treating cancer as demonstrated by previous unsuccessful clinical trials. Although multiple factors are responsible for poor clinical efficacy\, one major bottleneck is the reliance on FcγR-mediated crosslinking for sufficient receptor activation. This is inherently problematic because FcγR expression varies greatly on different immune cells\, leading to a wide range of receptor agonism. Emerging research suggests that antibodies engaging two different epitopes on the same immune receptor mediate receptor superclustering and enable robust antibody agonism without extrinsic Fc crosslinking. However\, there are no systematic methods for identifying such biepitopic (also known as biparatopic) agonist antibodies. Therefore\, the objective of this research work is to develop facile methods for reliably identifying biepitopic antibodies to activate immune receptors for immunotherapeutic applications.\n\nBiepitopic antibodies have been shown to mediate potent receptor activation for a variety of immune receptors. Traditionally\, the generation of these antibodies requires key steps including animal immunization\, epitope binning to identify unique antibody pairs\, and combining antibody pairs to engineer biepitopic antibodies. While this approach has been used to successfully discover biepitopic antibodies\, it suffers from key limitations. Notably\, animal immunization and subsequent antibody isolation is an arduous and unpredictable process. Even when successful clones are discovered from these processes\, further epitope binning experiments are needed to select antibody pairs to discover potent immune therapeutics. To overcome these limitations\, we developed an antibody screening strategy that greatly simplifies the discovery of biepitopic antibodies. Our approach eliminates the need for animal immunization by using existing\, off-the-shelf IgG antibodies specific to the target receptor. Next\, we perform in vitro selections by blocking the receptor epitope of the existing antibody and conducting subsequent sorts to identify single-chain antibodies with orthogonal binding domains. Thus far\, our work has shown that the antibody screening strategy can be used to discover antibodies for a variety of TNF receptors including OX40 and CD137.\n\nGiven that receptor clustering of three or more receptors is critical for activating TNF receptors\, we first generated biepitopic tetravalent OX40 antibodies by attaching novel single-chain antibodies to the C-termini of the light chain of existing clinical-stage antibodies. These tetravalent biepitopic antibodies showed remarkable T cell proliferation and cytokine secretion for biepitopic antibodies compared to their monoepitopic counterparts. Next\, we sought to improve the additional clinical-stage OX40 IgGs engineered as biepitopic antibodies to show the generality of our findings that biepitopic antibodies can mediate superior and FcγR-independent activities. Beyond OX40 IgGs\, we also show that biepitopic antibodies can be used to mediate superior T cell proliferation for other TNF receptors including CD137.
